摘要目的:探讨1,25二羟维生素D3及TGFβ/Smads信号通路与胃癌的关系。方法:收集93例胃癌组织和30例正常胃黏膜组织,采用免疫组化染色法检测1,25二羟维生素D3及TGFβ1、Smad2蛋白的表达。结果:胃癌组织中1,25二羟维生素D3表达水平低于正常胃黏膜组织(t=133.850,P<0.001),TGFβ1、Smad2表达水平高于正常胃黏膜组织(t=71.502、35.988,P<0.001)。1,25二羟维生素D3、TGFβ1和Smad2的表达水平与胃癌的分化程度、临床分期、浸润深度及淋巴结转移关系密切(P<0.001)。胃癌组织中1,25二羟维生素D3与TGFβ1、Smad2的表达呈负相关(r=-0.583,-0.462,P<0.001),TGFβ1与Smad2的表达呈正相关(r=0.425,P<0.001)。 结论:1,25二羟维生素D3可能通过调控TGFβ/Smads信号通路参与了胃癌的发生发展。
Abstract:
AbstractAim: To investigate the relationship between TGFβ/Smads signal pathway, 1,25dihydroxyvitamin D3 and gastric cancer. Methods:A total of 93 cases of gastric carcinoma specimens and 30 cases of normal gastric tissue were subjected to detect the expressions of 1,25dihydroxyvitamin D3, TGFβ1 and Smad2 by immunohistochemistry. Results: Compared with those of the normal gastric tissue, the expression of 1,25dihydroxyvitamin D3 was lower in gastric cancer tissue, and the expressions of TGFβ1 and Smad2 were higher(t=133.850,71.502,35.988,P<0.001). The expressions of 1,25dihydroxyvitamin D3, TGFβ1, and Smad2 had a close relationship with the degree of differentiation,clinical stage, depth of invasion and lymph node metastasis in gastric cancer tissue(P<0.001).The expression of 1,25dihydroxyvitamin D3 was negatively correlated with those of TGFβ1 and Smad2(r=-0.583,-0.462,P<0.001), and there was a positively correlation between TGFβ1 and Smad2 (r=0.425,P<0.001). Conclusion: 1,25dihydroxyvitamin D3 may be involved in gastric carcinogenesis and development by regulating TGFβ/Smad signal pathway.
