摘要目的:探讨子痫前期患者胎盘组织中DLX3和Syncytin的表达及意义。方法:选取子痫前期患者60例 (其中轻度子痫前期患者30例、重度子痫前期患者30例),正常妊娠妇女30例作为对照,在终止妊娠前3 d内应用彩色多普勒超声检测胎儿脐动脉血流S/D值与胎头双顶径,并且分别采用RTPCR和ELISA检测胎盘组织中DLX3和Syncytin mRNA、蛋白的表达。结果:①轻、重度子痫前期组DLX3及Syncytin mRNA的表达均明显低于对照组,且重度子痫前期组低于轻度子痫前期组 (F=694.115、546.194,P<0.05)。②轻、重度子痫前期组DLX3及Syncytin蛋白的表达均明显低于对照组,且重度子痫前期组低于轻度子痫前期组 (F=129.315、62.317,P<0.05)。③轻、重度子痫前期组中DLX3和Syncytin的表达均呈正相关(mRNA:r=0.429、0.513;蛋白:r=0.579、0.677,P<0.05)。④轻度子痫前期组DLX3和Syncytin蛋白的表达与胎儿脐动脉血流S/D值均呈负相关(r=-0.324和-0.352,P<0.05),与胎头双顶径呈正相关(r= 0.431和 0.510,P<0.05)。重度子痫前期组DLX3和Syncytin蛋白的表达与胎儿脐动脉血流S/D值均呈负相关(r=-0.624和-0.637,P<0.05),与胎头双顶径呈正相关(r=0.536和 0.617,P<0.05)。结论:DLX3和Syncytin在子痫前期的发病及疾病进展过程中起一定作用。
Abstract:
AbstractAim: To detect the expressions of homeobox gene DLX3 and Syncytin in placenta of patients with preeclampsia. Methods:A total of 60 women with preeclampsia(30 cases of mild preeclampsia and 30 cases of severe preeclampsia) and 30 normal pregnant women(control group) were recruited. Color Doppler ultrasound was used to detect fetal umbilical artery blood systolic/diastolic ratio and biparietal diameter within three days before the termination of pregnancy. DLX3, Syncytin mRNA and protein were detected by semiquantitative RTPCR and enzymelinked immunosorbent assay(ELISA). Results: The mRNA expressions of DLX3, Syncytin in mild and severe preeclampsia groups were both lower than those in control group,and those of the severe preeclampsia were lower than the mild preeclampsia(F=694.115,546.194,P<0.05). The protein expressions of DLX3, Syncytin in mild and severe preeclampsia group were both lower than the control group and those of the severe preeclampsia were lower than the mild preeclampsia(F=129.315,62.317,P<0.05). The mRNA and protein expressions of DLX3 and Syncytin in mild and severe preeclampsia groups were positively related(r= 0.429,0.513,0.579,0.677,P<0.05). The protein expressions of DLX3 and Syncytin were negatively related to umbilical artery blood systolic/diastolic ratio(r=-0.324,-0.352,P<0.05), and positively related to biparietal diameter (r= 0.431, 0.510,P<0.05)in mild preeclampsia group. The protein expressions of DLX3 and Syncytin were negatively related to umbilical artery blood systolic/diastolic ratio(r=-0.624,-0.637,P<0.05), and positively related to biparietal diameter in severe preeclampsia group(r= 0.536, 0.617,P<0.05).Conclusion: The low expressions of DLX3 and Syncytin play roles in pathogenesis and progress of preeclampsia.
