摘要:
目的:对豫医卷毛大鼠的表皮生长因子(EGF)基因进行比较生物学分析。方法:采用PCR方法对豫医卷毛大鼠EGF基因cDNA序列进行克隆测序,并与BN大鼠、家鼠、原鸡、猕猴和人EGF基因的核苷酸序列及其编码蛋白的氨基酸序列进行同源性比较和系统遗传学分析。结果:获得了豫医卷毛大鼠EGF基因全长3 522 bp的cDNA序列(GenBank登录号:JX437943)。豫医卷毛大鼠EGF基因编码区的核苷酸序列与BN大鼠、家鼠、原鸡、猕猴和人的同源性分别为97.8%、83.4%、59.7%、76.5%、73.5%,其编码蛋白的氨基酸序列同源性分别为99.3%、79.9%、52.9%、69.6%、69.4%。结论:EGF基因在进化过程中具有一定的保守性,但不同物种之间也具有特异性。
Abstract:
Aim: To study the difference of epidermal growth factor(EGF) gene and its coding protein between Yuyi curly hair rats and others species. Methods:PCR technique was developed to clone the sequence of cDNA of EGF gene from Yuyi curly hair rats, and biologic characters of nucleotide and amino acid sequence encoding EGF gene in different species (Norway rats, house mice, chicken, rhesus monkey, and human) were analyzed. Results: A fulllength cDNA of Yuyi curly hair rats was 3 522 bp (Accession Number: JX437943). The EGF gene nucleotide homology of Yuyi curly hair rats to Norway rats, house mice, chicken, rhesus monkey, and human was 97.8%, 83.4%, 59.7%, 76.5%, and 73.5%, and that of amino acid sequences was 99.3%,79.9%,52.9%,69.6%,and 69.4%, respectively. Conclusion: A certain degree of conservation of EGF gene exists among different mammalian taxons, but there were some specificity among different mammalian taxons.
