摘要:
目的:研究大鼠原位肺移植后体内CD8+调节性T细胞(CD8+ Treg)的分布及动态变化规律。方法:以cuff技术构建原位左肺移植的Wistar→Lewis大鼠同种肺移植模型20只、Lewis→Lewis同基因移植模型5只以及假手术模型5只,分别在术后不同时间点处死大鼠,切除移植物或者对照肺组织行常规病理学检查,以离心法、研磨法以及裂解法分别获取血液、脾脏、肺门淋巴结以及移植肺内的淋巴细胞,行流式细胞学检查。结果:同种移植组肺内可以见到典型的急性排斥反应。大鼠的血液和组织内检测到了CD8+CD45RC-Treg。同基因移植组肺组织及淋巴结内CD8+CD45RC-Treg/CD3+和CD8+CD45RC-Treg/CD8+分别为(15.88±2.72)%、(13.88±2.81)%和(30.68±5.25)%、(31.43±4.56)%,高于同种移植组(11.27±3.06)%、(9.89±1.86)%和(18.04±4.65)%、(18.89±4.79)%(t=2.518、2.648和4.030、4.240,P均<0.05)。结论:原位肺移植后肺组织及淋巴结内出现CD8+CD45RC-Treg的聚集,排斥反应发生后其在肺内的数量减少。
Abstract:
Aim: To study the dynamics and distribution of CD8+Treg in rat orthotopic lung transplant recipients. Methods:Allogeneic(n=20), isogeneic(n=5) orthotopic lung transplantation by cuff technique or sham operations(n=5) were performed for Lewis recipients. The rats were sacrificed at different postoperative time. Grafts or control lungs were harvested for histological evaluation. Lymphocytes in peripheral blood,spleen and local lymph nodes, and graft were acquired by centrifugation, trituration and lysis. Their phenotype and the expression of Foxp3 were analyzed by flow cytometry. Results: Typical acute rejections were observed in lung allografts. CD8+CD45RC-Treg was identified in blood and tissue. CD8+CD45RC-Treg/CD3+ and CD8+CD45RC-Treg/CD8+ in grafts and hilar lymph nodes of rats from isotransplant group were (15.88±2.72)%,(13.88±2.81)% and (30.68±5.25)%,(31.43±4.56)%, which were higher than those in allotransplant group [(11.27±3.06)%,(9.89±1.86)% and (18.04±4.65)%,(18.89±4.79)%](t=2.518, 2.648, 4.030, 4.240,P<0.05).Conclusion: CD8+CD45RC-Treg could aggregate to the grafts and local hilar lymph nodes. Lung Treg decrease in the setting of rejection.
