摘要:
目的:观察饮食干预对妊娠期大鼠胰岛素抵抗(IR)状态的影响。方法:雌性SD大鼠40只,随机分为高脂高糖饮食孕鼠组(SFP)、高脂高糖饮食孕后普食组(GHP)、普通饮食孕鼠组(NP)、空白对照组(NC),每组10只。SFP和GHP组给予高脂高糖饲料喂养,NP和NC组给予普通饲料喂养,6周后,SFP、GHP和NP组大鼠与雄性大鼠合笼受孕。SFP组继续给予高脂高糖饲料喂养,其他各组均给予普通饲料喂养。妊娠第20天,大鼠禁食12 h后,测体质量,计算妊娠前后体质量增量;心脏穿刺抽血测血浆空腹胰岛素(Fins)、空腹血糖(FBG)和血脂;行高胰岛素正糖钳夹实验,测葡萄糖输注率(GIR)。结果:4组大鼠FBG、Fins、GIR、体质量增量差异均有统计学意义(F=1 494.300,631.570,457.430和44.379,P<0.001)。SFP组大鼠GIR低于NP组(P<0.05),Fins和FBG高于NP组(P<0.05)。GHP组GIR较SFP组明显升高(P<0.05),Fins和FBG较SFP组明显降低(P<0.05)。结论:孕后饮食干预能明显改善高脂高糖诱导的妊娠期大鼠IR状态。
Abstract:
Aim: To evaluate the effect of dietary interventions on insulin resistance(IR) in pregnant rats. Methods:A total of 40 female SD rats were divided into four groups: SFP group, GHP group, NP group and NC group, 10 in each group. SFP and GHP groups were given high sucrose and fat diet,NP and NC groups were given normal diet. Six weeks later,the female rats of SFP,GHP and NP groups were put into the same cage with the male rats to conceive. At the 20th day,the weight were detected, and the weight gain were calculated, the levels of fast insulin (Fins),fast glucose(FBG),and blood lipids were detected,and hyperinsulinemiceuglycemic clamp test was performed to detect glucose infusion rate(GIR).Results: The weight gain,the levels of Fins and FBG,and GIR among the 4 groups had differences(F=44.379,631.570,1 494.300 and 457.430,P<0.001). Compared with those of the NP group, the levels of Fins and FBG of SFP group were higher, and that of GIR was lower(P<0.05). Compared with those of the SFP group, the levels of Fins and FBG of GHP group were lower, and that of GIR was higher(P<0.05). Conclusion: Dietary intervention could significantly improve the state of IR in pregnant rats with high fat and sugar diet.
