摘要:
目的:探讨活性维生素D3 对糖尿病大鼠肾脏骨形成蛋白7(BMP7)和转化生长因子β1(TGFβ1)表达的影响。 方法:30只雄性SD大鼠,按随机数字表法分3组:正常对照(NC)组,糖尿病模型(DM)组,活性维生素D3(DC)组(制备糖尿病模型后用骨化三醇灌胃),每组10只。12周后,检测大鼠24 h尿蛋白定量及血生化指标,采用PAS及Masson染色观察大鼠肾脏病理改变,免疫组化法检测肾组织中BMP7及TGFβ1蛋白的表达,RTPCR法检测肾组织中BMP7、TGFβ1 mRNA的表达。结果:与NC组比较,DM组大鼠肾脏肥大指数、24 h尿蛋白定量、血肌酐、尿素氮、胱抑素C水平均明显升高(P<0.05);同时肾组织中TGFβ1 mRNA及蛋白表达水平也明显升高(P<0.05),而BMP7 mRNA及蛋白表达水平明显下降(P<0.05)。与DM组比较,DC组肾组织中BMP7 mRNA及蛋白表达水平明显升高(P<0.05),余上述指标明显下降(P<0.05),病理改变明显减轻。结论:活性维生素D3可能通过上调BMP7、下调TGFβ1的表达,发挥其肾脏保护作用。
Abstract:
Aim: To explore the effect of 1, 25dihydroxyvitamin D3 on the expressions of bone morphogenetic protein7(BMP7) and transforming growth factorbeta 1(TGFβ1) in renal tissues of diabetic rats. Methods:A total of 30 SD rats were randomized into 3 groups: normal control(NC) group, diabetic model(DM) group and diabetic model treated with 1, 25dihydroxyvitamin D3(DC) group,and 10 in each group. After 12 weeks of treatment, the renal function, 24 hours total urine protein, blood calcium and so on were detected, PAS and Masson staining were performed to observe histological damage in the kidneys,and immunohistochemical staining and RTPCR were used to monitor the expressions of BMP7 and TGFβ1 protein and mRNA in renal tissue. Results: Compared with those of NC group, kidney index,24 hours total urine protein, Scr, serum BUN and CysC levels were significantly higher in DM group(P<0.05),and the mRNA and protein expressions of TGFβ1 in renal tissue were also higher(P<0.05),but the expressions of BMP7 were lower (P<0.05). Compared with those of DM group, the indexes mentioned above improved significantly(P<0.05),and the histological damage was alleviated.Conclusion: 1, 25dihydroxyvitamin D3 has some renal protective effect on diabetic rats, partly through upregulating BMP7 and downregulating TGFβ1 expressions in renal tissue.
