摘要:
目的:观察三七总皂苷对大鼠心肌梗死后血管新生和VEGF、微小RNA21(miRNA21)表达的影响。方法:108只健康Wistar大鼠结扎左冠状动脉建立急性心肌梗死模型后分为心肌梗死对照组、三七总皂苷低剂量治疗组和三七总皂苷高剂量治疗组,心肌梗死对照组和三七总皂苷治疗组分别于术后即刻及术后每天腹腔注射生理盐水和三七总皂苷(100和400 mg/kg),术后3、7、14、21 d分别处死9只大鼠,进行心肌梗死面积百分比测定,免疫组化法测定梗死周边区微血管密度和VEGF蛋白相对表达量,RTPCR法检测miRNA21、VEGF mRNA相对表达量。结果:3组3、7、14及21 d梗死周边区微血管密度、心肌梗死面积百分比、VEGF蛋白相对表达量、VEGF和miRNA21 mRNA相对表达量比较,差异均有统计学意义(F微血管密度=35.163、42.906、49.510、54.245,P均<0.001;F梗死面积百分比=59.358、68.241、66.350、58.667,P均<0.001;FVEGF 蛋白=145.576、164.024、179.349、186.362,P均<0.001; FVEGF mRNA=253.542、268.958、315.313、347.914,P均<0.001; FmiRNA21 mRNA=53.421、67.954、72.358、75.694,P均<0.001)。与心肌梗死对照组相比,三七总皂苷治疗组微血管密度、VEGF蛋白、VEGF和miRNA21 mRNA相对表达量均升高(P均<0.05),心肌梗死面积百分比减少(P均<0.05);不同剂量三七总皂苷治疗组上述各指标比较,差异均有统计学意义(P均<0.05)。各组梗死周边区miRNA21、VEGF mRNA的相对表达量间均呈正相关(r心肌梗死对照组=0.609,P=0.036;r三七总皂苷低剂量治疗组=0.522,P=0.047;r三七总皂苷高剂量治疗组=0.581,P=0.042)。结论:三七总皂苷有促进心肌梗死后VEGF和miRNA21表达、促进血管新生和缩小梗死面积的作用。
Abstract:
Aim: To explore the effects of panax notoginseng saponins(PNS)on angiogenesis and VEGF, miRNA21 expressions in rats with myocardial infarction.Methods: The left coronary artery of 108 male Wistar rats was ligated to make the models of acute myocardial infarction.All rats were randomly divided into myocardial infarction control group,low PNS group(100 mg/kg)and high PNS group(400 mg/kg). The microvascular density in the infarct area, VEGF protein, miRNA21 and VEGF mRNA were detected at 3,7,14,21 day after operation.Results: At 3, 7, 14 and 21 day,there were significant differences in microvascular density,infarct areas,the expressions of VEGF protein,VEGF and miRNA21 mRNA among the three groups(Fmicrovascular density=35.163, 42.906, 49.510, 54.245,P<0.001;Finfarct areas=59.358, 68.241, 66.350, 58.667,P<0.001;FVEGF protein=145.576,164.024,179.349,186.362,P<0.001; FVEGF mRNA=253.542, 268.958, 315.313, 347.914,P<0.001; FmiRNA21 mRNA=53.421, 67.954, 72.358, 75.694,P<0.001).The infarct areas decreased significantly in PNS groups while the other 3 indexes increased compared with myocardial infarction control group(P<0.05).There were also significant differences in the above indexes between low PNS group and high PNS group(P<0.05).The expressions of miRNA21 and VEGF mRNA in infarct areas had positive correlation(r myocardial infarction control group=0.609,P=0.036;Flow PNS group=0.522,P=0.047;Fhigh PNS group=0.581,P=0.042).Conclusion: PNS could increase VEGF and miRNA21 expression, improve angiogenesis and decrease infarct area after myocardial infarction.
