摘要:
目的:观察不同含量常压氧(NBO)对急性脑缺血再灌注大鼠脑组织Caspase9及Caspase3 mRNA表达的影响。 方法:选取制作脑缺血再灌注损伤模型成功的120只SPF级雄性SD大鼠,分为低、中、高含量NBO组和空气组,在持续吸氧或空气3、6、12、24、48、72 h时,各组分别随机抽取5只大鼠处死,断头取脑,用荧光定量PCR检测Caspase9及Caspase3 mRNA的表达水平。结果:空气组和低、中、高含量NBO组Caspase9 及Caspase3 mRNA的表达在48 h达到高峰,之后开始下降(P均<0.05),且各时间点NBO组均低于空气组(P均<0.05)。结论:NBO可以下调急性脑缺血再灌注后脑组织Caspase9及Caspase3 mRNA的表达。
Abstract:
Aim: To explore the effects of different concentrations of normobaric oxygen(NBO) on Caspase9 and Caspase3 mRNA expression in brain tissue of rats after acute cerebral ischemiareperfusion. Methods: A total of 120 SPF level male SD rats were subjected to middle cerebral artery occlusionreperfusion model. The rats were randomly divided into four groups, of which 3 groups (n=90) were given three concentrations of NBO treatment respectively and the other group was the air group (n=30). When taking persistent oxygen or air for 3, 6, 12, 24, 48, 72 h, five rats randomly taken in each group would be sacrificed, with brain collected. Fluorescence quantitative PCR was used to detect Caspase9 and Caspase3 mRNA expression levels. Results: The expression of Caspase9 and Caspase3 mRNA peaked in air group and low, medium, high concentration of NBO treatment groups at 48 h, then began to decline(P<0.05), and at each time point those of the NBO groups were lower than that of air group(P<0.05). Conclusion: NBO could reduce Caspase9 and Caspase3 mRNA expression in cerebral ischemiareperfusion.
