目的:探讨结肠癌组织中SLP2蛋白的表达情况及SLP2基因对结肠癌细胞HCT116侵袭、迁移力的影响。方法:采用免疫组化法检测50对结肠癌及正常结肠黏膜组织中SLP2蛋白的表达,分析患者临床病理特征与SLP2蛋白表达的关系。设计合成SLP2 siRNA,转染结肠癌HCT116细胞,Transwell法及Matrigel基质实验检测转染SLP2 siRNA对HCT116细胞侵袭、迁移力的影响,RTPCR及Western blot检测转染48 h后HCT116细胞中SLP2 mRNA及蛋白的表达情况。结果:正常结肠及结肠癌组织中SLP2蛋白的高表达率比较,差异有统计学意义(χ2配对=23.188,P<0.001)。SLP2蛋白表达与结肠癌TNM分期(χ2=10.474,P=0.001)及淋巴结转移(χ2=19.647,P<0.001)有关。SLP2 siRNA转染组细胞迁移力、侵袭力、SLP2 mRNA及蛋白表达水平均低于阴性对照组和空白对照组(P均<0.05)。结论:结肠癌组织中SLP2高表达;SLP2可能通过促进结肠癌细胞的侵袭、迁移能力,参与结肠癌的转移。
Abstract:
Aim: To investigate SLP2 expression in colorectal cancer tissue and its influence on HCT116 cell's migration and invasion. Methods: Immunohistochemistry was used to detect the expression of SLP2 protein in 50 normal colorectal tissue and colorectal cancer tissue. Then the relationship between the expression of SLP2 protein and the clinical pathological characteristics of the 50 colorectal cancer cases was analyzed. SLP2 siRNA was transfected into colorectal cancer cell line HCT116. The SLP2 mRNA and protein expression in HCT116 cells were detected using RTPCR and Western blot. The cell migration and invasion ability was detected by Transwell test and Matrigel invasion test. Results: The high SLP2 expression rate in coloretal cancer tissue was higher than that of the adjacent normal colorectal tissue(χ2paired= 23.188,P<0.001). The expression of SLP2 in coloretal cancer tissue was associated with TNM staging(χ2=10.474,P=0.001) and lymph nodes metastasis (χ2=19.647,P<0.001). After interfering with SLP2 siRNA, the migration and invasion ability of HCT116 cell decreased, and the exprssion of SLP2 mRNA and protein decreased(P<0.05). Conclusion: SLP2 protein shows upregulation in colorectal cancer, and it may promote cell metastasis through promoting migration and invasion ability.
