目的:探讨姜黄素(Cur)对多囊卵巢综合征(PCOS)大鼠代谢紊乱的影响。方法:30只雌性SD大鼠随机分为正常组、模型组和Cur组,每组10只。模型组和Cur组通过灌胃来曲唑建立PCOS模型。Cur组大鼠每日灌胃Cur(200 mg/kg)0.5 mL,模型组和正常组大鼠灌胃等体积的生理盐水,连续14 d 。测定大鼠体重、双侧卵巢质量、空腹血糖(FBG),放射免疫法测定血清黄体生成素(LH)、睾酮(T)、空腹胰岛素(FINS),酶联免疫法测定血清脂联素(APN),计算稳态模型胰岛素抵抗(HOMAIR)。结果:3组大鼠体重、双侧卵巢质量、LH、T、FBG、FINS、HOMAIR和APN比较差异有统计学意义(F=6.530、6.755、20.379、37.488、8.456、12.851、14.854和36.371 ,P<0.05);Cur组大鼠血清APN水平较模型组上升,其他指标较模型组降低(P<0.05)。结论:Cur能够降低PCOS大鼠雄激素水平及卵巢质量,可能与其可减轻大鼠体重、缓解胰岛素抵抗、调节脂联素水平有关。
Abstract:
Aim: To explore the effects of curcumin (Cur) on rats with polycystic ovary syndrome (PCOS). Methods: 30 female SD rats were randomly divided into normal group, model group, Cur (200 mg·kg-1) group, and 10 in each group. The model of PCOS was set up by intragastricly administrating with letrozole. Body mass, bilateral ovarian weight, fasting blood glucose (FBG) were determined, luteinizing hormone (LH), testosterone (T), insulin (FINS) in serum were assayed with radioimmunoassay method, adiponectin (APN) in serum was detected with enzymelinked immunosorbent assay method and homeostasis model assessment of insulin resistance (HOMAIR) was calculated. Results: The body weight, bilateral ovarian mass, LH, T, FBG, FINS, HOMAIR and APN had significant differences (F=6.530,6.755,20.379,37.488,8.456,12.851,14.854 and 36.371,P<0.05); APN level in Cur group significantly increased as compared with the model group,and the other indexes decreased (P<0.05). Conclusion: Curcumin can decrease androgen level and ovarian weight of PCOS rats, which may be related with its reducing weight, relieving insulin resistance and regulating adiponectin.
