摘要目的:对比不同倍比稀释条件稀释血清对快速荧光灶抑制试验(RFFIT)结果的影响,以指导RFFIT在狂犬病防控中的应用。方法:采集65个健康个体和40个狂犬病暴露后预防处置(PEP)个体的血清标本,分别进行3倍和5倍倍比稀释,采用RFFIT检测抗狂犬病病毒中和抗体(RVNA)水平。结果:3倍倍比稀释时,RVNA检测结果显示:65个健康个体中,63人<0.50 IU/mL,2人≥0.50 IU/mL;40个PEP个体均≥0.50 IU/mL。5倍倍比稀释时,RVNA检测结果显示:65个健康个体均<0.50 IU/mL;40个PEP个体中,34例≥0.50 IU/mL,6例<0.50 IU/mL,这6例PEP个体在3倍倍比稀释时RVNA呈弱阳性,效价在0.50~1.00 IU/mL。结论:以5倍倍比稀释条件稀释血清进行RFFIT检测时灵敏度降低,但是检测结果更能反映个体实际免疫状态,可能更有利于指导狂犬病PEP措施。
Abstract:
AbstractAim: To compare the effects of different diluting conditions of sera on the results of rapid fluorescent focus inhibition test(RFFIT), so as to direct the application of RFFIT in rabies control and prevention. Methods: Sera samples from 65 healthy persons and 40 rabies postexposure prophylaxis(PEP) individuals were collected. The sera were diluted by 13 and 15 coubling dilution respectively and the rabies virus neutralizing antibody(RVNA) was detected by RFFIT.Results: With 13 coubling dilution of sera, RVNA results indicated that among 65 healthy persons, 63<0.50 IU/mL but 2≥0.50 IU/mL; all of the 40 PEP individuals ≥0.50 IU/mL. With 15 coubling dilution of sera, RVNA results indicated that all of 65 healthy persons <0.50 IU/mL; among 40 PEP individuals, 34≥0.50 IU/mL, 6<0.50 IU/mL, who had weak positive results (0.50~1.00 IU/mL) with 13 dilution. Conclusion: With 15 dilution of sera, the sensitivity of RFFIT is decreased, but the RVNA results reflect the real immunologic status more actually, which should be helpful to direct the PEP measurements.
