摘要:
目的:通过单细胞凝胶电泳技术探讨臭氧急性暴露对BALB/c小鼠肺细胞DNA的断裂作用。方法:12只SPF级雄性BALB/c小鼠分为阴性对照组和臭氧暴露组,每组6只。臭氧暴露组小鼠用体积分数0.000 1%的臭氧在动态染毒控制系统中染毒3 h,阴性对照组置于同一动态染毒柜中,通入过滤后的空气(不含臭氧)。染毒结束24 h后取肺细胞,应用单细胞凝胶电泳技术检测肺细胞DNA的断裂程度。结果:阴性对照组肺细胞的尾部DNA含量百分比、尾矩和Olive尾矩分别为1.153%(0.086%,5.324%)、0.126(0.004,0.873) μm和0.499(0.052,2.347) μm,臭氧暴露组肺细胞的尾部DNA含量百分比、尾矩和Olive尾矩分别为29.669%(22.541%,37.621%)、22.456(13.464,38.596) μm和15.159(10.275,25.966) μm。臭氧暴露组较阴性对照组各指标均升高(Z=13.295、13.233、12.984,P均<0.001)。结论:体积分数0.000 1%臭氧急性暴露3 h会导致BALB/c小鼠肺细胞DNA断裂损伤。
Abstract:
Aim: To explore shortterm exposure to ozone induced DNA strand break of lung cells in BALB/c mice by single cell gel electrophoresis. Methods: Twelve SPF grade BALB/c mice were randomly divided into control group and ozone exposure group,each group of 6 mice. The mice of ozone exposure group were exposed to 0.000 1% of ozone for 3 hours in a wholebody exposure chamber and control mice were exposed to filtered air without ozone. After exposure, lung cells were taken rapidly. DNA damage of lung cells was tested using single cell gel electrophoresis. Results: The tail DNA percentage, tailmoment and Olive tailmoment of the control group were respectively 1.153%(0.086%,5.324%), 0.126(0.004,0.873) μm, and 0.499(0.052,2.347) μm. The tail DNA percentage, tailmoment and Olive tailmoment of ozone exposure group were respectively 29.669%(22.541%, 37.621%), 22.456(13.464, 38.596) μm, and 15.159(10.275,25.966) μm.The differences were significant between the two groups(Z=13.295, 13.233, and 12.984, P<0.001). Conclusion: Shortterm 0.000 1% ozone exposure for 3 hours could cause DNA strand break in BALB/c mice lung cells.
