摘要:
目的:探讨微小RNA146a(miR146a)在M1型、M2型巨噬细胞及肿瘤相关巨噬细胞(TAMs)中表达的差异。方法:将单核细胞系(THP1)细胞诱导为M1型、M2型巨噬细胞及TAMs,应用流式细胞仪检测表型后,采用实时荧光定量PCR方法分别检测THP1、M1型和M2型巨噬细胞及TAMs中miR146a的表达情况。结果:THP1细胞中miR146a的表达量为(0.838±0.030),M1型巨噬细胞中的表达量为(1.788±0.384),M2型巨噬细胞中的表达量为(2.760±0.561),TAMs中的表达量为(2.974±0.661),差异有统计学意义(F=12.847,P=0.002)。其中M1型(P=0.040)、M2型(P=0.001)巨噬细胞及TAMs(P=0.001)中miR146a的表达量高于THP1细胞,TAMs(P=0.015)与M2型巨噬细胞(P=0.036)中的miR146a表达量高于M1型巨噬细胞,TAMs与M2型巨噬细胞中miR146a的表达差异无统计学意义(P=0.595)。结论:miR146a在M1型、M2型巨噬细胞中的差异性表达可能与其分化和(或)细胞功能相关;TAMs的表型特征可能与M2型巨噬细胞类似,同时可能具有与M2型巨噬细胞相同的细胞功能。
Abstract:
Aim: To investigate the expression of miRNA146a in different phenotypes of activated macrophages.Methods: In this study,the THP1 induced into M1type,M2type macrophages or tumorassociated macrophages(TAMs),and their phenotypes were confirmed by flow cytometry. The miRNA146a expression was determined by qRTPCR in M1type macrophages,M2type macrophages and TAMs. Results: The miRNA146a expression was significantly increased in M1type macrophages [(1.788±0.384), P=0.040], M2type macrophages [(2.760±0.561), P=0.001]and TAMs [(2.974±0.661),P=0.001] compared with the THP1 (0.838±0.030); The miRNA146a in THP1,M1type, M2type macrophages and TAMs had significant difference(F=12.847,P=0.002). The miR146a expression was significantly increased in the M2type macrophages(P=0.036)and the TAMs(P=0.015) compared with the M1type macrophages. There was no statistical significance between TAMs and M2type macrophages in the expression of miRNA146a(P=0.595).Conclusion: Different expressions of miRNA146a in M1type and M2type macrophages may associate with the differentiation or their cellular functions. The phenotypic characteristics of TAMs and M2type macrophages are similar,and they may have the same functions.
