摘要:
目的:观察羊膜对在其基质面生长的人子宫内膜细胞肝细胞生长因子(HGF)、基质金属蛋白酶9(MMP9)及血管内皮生长因子(VEGF)表达的影响。方法:将原代培养正常人子宫内膜细胞接种于羊膜的基质面作为羊膜载体组,以无羊膜载体培养的子宫内膜细胞为对照组。培养8 d后,实时荧光定量PCR法检测2组细胞HGF、MMP9、VEGF mRNA表达的变化,ELISA法检测2组细胞上清液MMP9蛋白含量。结果:羊膜载体组子宫内膜细胞HGF mRNA相对表达量为(2.033±0.922),MMP9 mRNA相对表达量为(1.356±0.399),VEGF mRNA相对表达量为(0.976±0.597);对照组分别为(1.299±1.003)、(0.871±0.529)、(0.426±0.193)(t=3.061、2.613、3.313,P均<0.05)。ELISA法检测细胞上清液MMP9蛋白含量,羊膜载体组为(0.762±0.085) μg/L,对照组为(0.089±0.008) μg/L(t=29.370,P<0.001)。结论:羊膜载体可以增强人子宫内膜细胞HGF、VEGF、MMP9的表达。
Abstract:
Aim: To observe the expressions of HGF, MMP9 and VEGF in human endometrial cells on the stromal surface of amniotic membrane. Methods: Primary cultured normal human endometrial cells were seeded on the stromal surface of amniotic membrane as the experimental group, no carrier in cultured amniotic endometrial cells as the control group. HGF, MMP9, and VEGF mRNA expression changes were detected by fluorescence quantitative PCR, and ELISA was used to detect MMP9 protein in supernatant fluid. Results: In the experimental group, HGF mRNA was (2.033±0.922), MMP9 mRNA was (1.356±0.399),VEGF mRNA was (0.976±0.597). In the control group, HGF mRNA was (1.299±1.003),MMP9 mRNA was (0.871±0.529),VEGF mRNA was (0.426±0.193). The differences were significant(t=3.061,2.613,3.313,P<0.05). MMP9 protein expression in cell supernatant fluid of experimental group was (0.762±0.085) μg/L, and higher than that of control group[(0.089±0.008) μg/L] (t=29.370,P<0.001). Conclusion: Amniotic membrane carrier could enhance expressions of HGF, VEGF, and MMP9 in human endometrial cells.
