摘要:
目的:探讨卵巢上皮性癌组织中PITX2和βcatenin蛋白的表达及意义。方法:采用免疫组化法检测80例卵巢上皮性癌组织、40例卵巢上皮性良性肿瘤组织和40例正常卵巢组织中PITX2与βcatenin蛋白的表达。采用ELISA检测40例卵巢上皮性癌患者术前和术后(7 d)血清中PITX2蛋白的含量。结果:不同类型卵巢组织中PITX2、βcatenin蛋白的表达差异有统计学意义(χ2=85.380和80.200,P<0.001),卵巢上皮性癌组织中PITX2蛋白的阳性表达率与βcatenin蛋白的异位阳性表达率高于卵巢上皮性良性肿瘤组织和正常卵巢组织,而两者在卵巢上皮性良性肿瘤组织和正常卵巢组织中低表达或不表达。PITX2蛋白的阳性表达率及βcatenin蛋白的异位阳性表达率与卵巢上皮性癌的组织分化程度、FIGO分期、淋巴结转移有关(P<0.05)。卵巢上皮性癌组织中PITX2表达与βcatenin的异位表达呈正关联(rP=0.411,P<0.001)。卵巢上皮性癌患者术前血清中PITX2的含量高于术后血清中的含量(t配对=16.582,P<0.001)。结论:卵巢上皮性癌组织中βcatenin的激活可能引起细胞中PITX2的上调,从而进一步引起细胞异常增殖、分化及肿瘤的发生发展。
Abstract:
Aim: To investigate the expressions and significance of PITX2 and βcatenin in epithelial ovarian cancer tissue. Methods: Immunohistochemical staining (SP method) was used to detect the expressions of PITX2 and βcatenin in 80 cases of epithelial ovarian cancer tissue,40 cases of benign epithelial ovarian neoplasm tissue,and 40 cases of normal epithelial ovarian tissue. ELISA was used to detect the expression of PITX2 in 40 cases of ovarian cancer blood serum obtained before operation and 7 days after operation. Results: The expressions of PITX2 and βcatenin had significant difference in different ovarian tissue(χ2=85.380,80.200,P<0.001).PITX2 protein expression and βcatenin protein ectopic expression in epithelial ovarian cancer tissue were significantly higher than those in benign ovarian tissue and normal ovarian tissue,but the expression of PITX2 and βcatenin were none or low in the benign ovarian tissue and normal ovarian tissue. The expression levels of PITX2 and βcatenin were associated with histological differentiation, FIGO staging and lymph node metastasis (P<0.05).PITX2 expression and βcatenin ectopic expression in epithelial ovarian carcinoma tissue were positively correlated (rP=0.411, P<0.001). The serum levels of PITX2 preoperation were significantly higher than that after operation(tpaired=16.582,P<0.001). Conclusion: The abnormal activation of βcatenin in ovarian epithelial carcinoma possibly activates its downstream target gene of PITX2,which leads to cell differentiation,proliferation and the occurrence and development of tumors.
