摘要:
目的:探讨二甲双胍对IL6诱导的胃癌SGC7901细胞上皮间质转化(EMT)的影响。方法:采用IL6培养SGC7901,构建EMT模型,倒置显微镜下观察细胞形态学变化。将SGC7901细胞分为对照组、IL6(50 μg/L)培养组、二甲双胍(50 mmol/L)培养组和二甲双胍+IL6培养组;培养48 h,MTT法检测各组细胞相对存活率,流式细胞术检测各组细胞周期的变化,细胞划痕修复实验检测细胞增殖及迁移的变化,RTPCR及Western blot检测细胞中Ecadherin、Ncadherin、Vimentin、Twist1 mRNA和蛋白表达的变化。结果:IL6能诱导SGC7901细胞由上皮细胞样形态向间质细胞样形态转化,多数细胞表现为似成纤维状,部分细胞呈梭形、纺锤体形。二甲双胍干预能抑制胃癌细胞的增殖(F=137.445,P<0.001)、细胞的周期分布及细胞的迁移和侵袭能力。二甲双胍干预后SGC7901细胞中Ecadherin mRNA和蛋白表达水平明显升高(F=34.678、53.190;P<0.001),而Ncadherin、Vimentin、Twist1 mRNA和蛋白表达水平降低(F=27.856、3.667、7.180及4.940、7.721、76.193;P均<0.05)。结论:二甲双胍能明显抑制胃癌细胞的增殖、侵袭及EMT过程。
Abstract:
To investigate the effect of metformin on epithelial mesenchymal transformation(EMT) of human gastric adenocarcinoma cell line SGC7901.Methods: SGC7901 cells were cultured with DMEM medium containing IL6(50 μg/L) to construct the EMT model.The morphological changes of the cells were observed with invert phase contrast microscope.According to the purpose of the experiment, SGC7901 cells were allocated into 4 groups,control group,metformin group,IL6 group and L6+metformin group. The survival rates of the cells were measured by MTT, cell cycle was analyzed by flow cytometry,the levels of Ecadherin, Ncadherin, Vimentin and Twist1 mRNA and protein in SGC7901 cells were determined by RTPCR and Western blot. Results: SGC7901 cells acquired more typical spindleshape phenotype of mesenchymal cells after cocultured with IL6,most of them presented fibroid form.The proliferation of SGC7901 cells was inhibited by metformin significantly(F=137.445,P<0.001). Metformin could affect the cell cycle distribution of SGC7901 cells and inhibit migration and invasion ability obviously.RTPCR and Western blot indicated that the mRNA and protein expression levels of Ecadherin were increased significantly(F=34.678,53.190;P<0.001),while the mRNA and protein expression levels of Ncadherin, Vimentin and Twist1 were obviously decreased via the effect of metformin(F=27.856,3.667,7.180 and 4.940, 7.721, 76.193;all P<0.05).Conclusion: Metformin can obviously inhibit proliferation,invasion ability and EMT process of gastric cancer cell.
