摘要:
目的:探讨牙周病大鼠不同正畸条件下牙根吸收情况与牙组织中基质金属蛋白酶9(MMP9)表达水平的变化。方法:取108只大鼠,54只制作大鼠牙周病模型,模型制备成功后分别给予0、50和80 g正畸力牵拉1、7、14 d,每个条件下6只大鼠;另54只给予相同正畸处理但不制备牙周病模型(对照组)。以上颌切牙为支抗,分别牵拉左侧上颌第一磨牙向近中移动。取各组大鼠实验牙制作组织切片,观察牙根吸收情况,应用免疫组化染色观察MMP9的表达。结果:50或80 g正畸力牵拉7 d后,两组大鼠牙周膜均排列紊乱,可见明显的牙根吸收现象,牙周病组大鼠牙根吸收情况较对照组严重;牵拉14 d后牙根吸收活动基本停止。随着正畸力的增加,MMP9表达水平逐渐升高;牵拉7 d后MMP9表达水平最高,14 d后下降;牙周病组牙组织中MMP9表达水平高于对照组(F组别=3.031, P=0.022;F正畸力=6.634,P=0.014;F时间=4.820,P=0.009;F交互=0.890,P=0.514)。结论:牙组织中MMP9表达水平可能能反映牙周病大鼠牙根吸收的程度。
Abstract:
To investigate the expression of MMP9 and root resorption by orthodontic force in rats with periodontal disease. Methods: A total of 108 rats were collected,among which 54 rats were made periodontal disease model,and then giving 0,50 or 80 g orthodontic force for 1,7,14 days,respectively,6 rats at each orthodontic condition;the other 54 rats(the control) were healthy and recepted the same orthodontic treatment. The histological slices of maxillary first molar were made to observe the histological changes of the root resorption and immunohistologic staining method was used to detect the expression of MMP9.Results: When being loaded orthodontic force of 50 or 80 g for 7 days, the rats in the 2 groups showed that periodontal ligament arranged in disorder, obvious root resorption, and the root resorption of the periodontal disease rats was more severe than the control; after 14 days,root absorption activities stopped. With the increase of orthodontic force, the expression of MMP9 increased; the expression of MMP9 on the 7th day was the highest, and decreased after the 14th day; the expression of MMP9 in periodontal disease group was higher than that in the control(Fgroup=3.031,P=0.022; Fforce=6.634,P=0.014;Ftime=4.820,P=0.009;Finteraction=0.890,P=0.514).Conclusion: The expression of MMP9 in tooth tissue may reflect the severity of root resorption of periodontal disease rats.
