摘要:
目的:通过观察miRNA let7b修饰的骨髓间充质干细胞(BMSCs)移植对脊髓损伤大鼠的运动功能恢复的作用,探讨miRNA let7b在损伤脊髓修复中的作用和机制。方法:60只成年SD大鼠分为对照组、BMSCs组、miRNA组,每组20只。采用改良Allen′s法制备脊髓损伤模型。造模后1周, BMSCs组行BMSCs移植,miRNA组移植miRNA let7b慢病毒载体感染的BMSCs,对照组注射生理盐水。移植后第3天至8周,按BBB标准量表对大鼠后肢运动功能进行评分,用免疫组化方法检测脊髓中神经丝蛋白200(NF200)和胶质纤维酸性蛋白(GFAP)的表达。结果:实验时间内,各组大鼠BBB评分和脊髓NF200表达持续升高;不同时间点BBB评分和脊髓NF200表达以miRNA组最高,BMSCs组次之,对照组最低(P<0.05)。移植后各组脊髓GFAP表达均呈持续性增高,各时间点GFAP阳性表达面积以对照组最大,BMSCs次之,miRNA组最小(P<0.05)。结论:miRNA let7b可能通过促进BMSCs分化为神经元样细胞、抑制脊髓损伤后反应性胶质细胞的增生及胶质瘢痕形成等机制促进损伤后的神经功能恢复。
Abstract:
To explore the effects of miRNA let7bmodified BMSCs transplantation on the repair of nerve in rats with spinal cord injury.Methods: A total of 60 SD rats were randomly divided into 3 groups:control group,BMSCs group, and miRNA group, each group including 20 rats. At 1 week after the spinal cord injury model being established, the control group received physical saline, the BMSCs group was injected with BMSCs,and the miRNA group was treated with BMSCs transfected with miRNA let7b. The neurological functions were evaluated using the BBB scale after operation.NF200 and GFAP were detected.Results: After cell transplantation, the BBB scores were the lowest in control group and lower in BMSCs group(P<0.05). After implantation,NF200positive cells of miRNA group was much more than the other two groups, while the GFAP positive area was the lowest in miRNA group and lower in BMSCs group(P<0.05).Conclusion: miRNA let7b may play an important role in the differentiation of BMSCs into neurons, and promote recovery of injured spinal cord through decreasing the formation of glial scar.
