目的:观察阿托伐他汀对心肌梗死大鼠心室重构和miRNA21表达的影响。方法:将48只健康Wistar大鼠随机分为假手术组、模型组、阿托伐他汀低剂量治疗组和高剂量治疗组,后3组结扎左冠状动脉前降支建立急性心肌梗死(AMI)模型,阿托伐他汀低剂量和高剂量治疗组分别按20和40 mg/(kg·d)的剂量灌胃给药。6周后测定左室质量指数(LVMI)、心肌胶原容积分数(CVF),RTPCR法检测梗死周边区心肌组织miRNA21及MMP9 mRNA的表达。结果:4组大鼠LVMI、心肌CVF及梗死周边区心肌组织中miRNA21和MMP9 mRNA的表达水平差异均有统计学意义(F=53.021、54.003、42.008和45.002,P均<0.05)。与模型组相比,阿托伐他汀治疗组上述指标均降低(P<0.05),低剂量和高剂量治疗组间上述指标的差异无统计学意义(P>0.05)。模型组、阿托伐他汀低剂量和高剂量治疗组梗死周边区心肌组织miRNA21表达水平与MMP9 mRNA水平呈正相关(r=0.613、0.512、0.551,P<0.05)。结论:阿托伐他汀能降低梗死周边区心肌组织MMP9 mRNA和miRNA21的表达,从而发挥缩小梗死面积、减轻心室重构的作用。
Abstract:
Aim: To explore the effect of atorvastatin on ventricular remodeling and expression of miRNA21 in rats with myocardial infarction. Methods: A total of 48 male Wistar rats were randomly allocated into 4 groups:sham operation group, model group, lowdose atorvastatin and highdose atorvastatin group,12 rats in each group.The rats in the latter 3 groups were made the acute myocardial infarction(AMI) model by ligating the left coronary artery. Lowdose atorvastatin and highdose atorvastatin groups were given atorvastatin at the doses of 20 and 40 mg/(kg·d) for 6 weeks,respectively. Then,left ventricular mass index(LVMI),collagen volume fraction(CVF),and the expressions of MMP9 mRNA and miRNA21 in the surrounding infarct areas were detected. Results: There were significant differences in LVMI, CVF, the expressions of miRNA21 and MMP9 mRNA among the four groups(F=53.021,54.003,42.008 and 45.002,P<0.05). LVMI, CVF,and the expressions of miRNA21 and MMP9 mRNA were significantly decreased in atorvastatin groups compared with those of the model group(P<0.05),while no significant differences were found between lowdose and highdose atorvastatin groups(P>0.05).The expression of miRNA21 was positively correlated with MMP9 mRNA in the model, lowdose and highdose atorvastatin group(r=0.613,0.512,0.551,P<0.05). Conclusion: Atorvastatin could decrease the expressions of MMP9 mRNA and miRNA21 in rats with AMI, which could decrease infarct area and improve ventricular remodeling and heart function.
