摘要:
目的:探讨培哚普利对盐酸异丙肾上腺素(ISO)所致心肌肥厚大鼠心肌组织中肌细胞增强因子2A(MEF2A)表达的影响。方法:30只雄性Wistar大鼠随机分为对照组、模型组、药物干预组,每组10只。对照组大鼠皮下注射生理盐水;模型组大鼠皮下注射ISO;药物干预组大鼠皮下注射ISO,同时用培哚普利进行灌胃。10 d后解剖心脏,计算全心重量/体重比及左心室重量/体重比;HE染色观察心肌病理变化;应用RTPCR及免疫组化方法检测心肌组织中MEF2A mRNA及蛋白的表达。结果:4个检测指标结果均显示模型组>药物干预组>对照组,差异均有统计学意义(P均<0.05)。结论:MEF2A参与心肌肥厚的发生发展;培哚普利可抑制心肌肥厚,其机制与调节MEF2A的表达有关。
Abstract:
Aim: To explore the effects of perindopril on expression of myocyte enhancer factor 2A(MEF2A) in myocardial tissue of myocardial hypertrophy rats induced by isoproterenol.Methods: A total of 30 male Wistar rats were randomly divided into control group, model group and drug intervention group(n=10). In control group physiological saline was subcutaneously injected,in model group isoproterenol was subcutaneously injected, in drug intervention group isoproterenol was subcutaneously injected and perindopril was gavaged.Ten days later,heart weight/body weight and left ventricle weight/body weight were calculated,and MEF2A mRNA and protein were detected by RTPCR and SP method. Results: The four indexes in model group were the highest,and were the lowest in control group(P<0.05).Conclusion: MEF2A promotes myocardial hypertrophy. Perindopril regulates the expression of MEF2A,which could inhibit myocardial hypertrophy.
