摘要:
目的:研究结直肠癌组织中miRNA表达谱的差异及其相关信号通路。方法:采用SOLiD高通量测序技术对10例结直肠癌组织和10例癌旁正常组织进行测序,然后利用生物信息学方法分析miRNA-mRNA相互作用的通路。结果:筛选出194个结直肠癌差异表达的miRNA,这些miRNA广泛参与基因表达的转录后调控,如MAPK、WNT、TGF-β信号通路。结论:结直肠癌miRNA的差异表达可能暗示着有不同功能或来源的miRNA通过调控不同的靶基因来协同、精细调控某些特定的信号通路。
Abstract:
Aim: To investigate the differences of miRNA expression profile and related signaling pathways in colorectal cancer.Methods: SOLiD high-throughput sequencing technology was used for investigating differentially expressed miRNA in the 10 paired colorectal samples(10 colorectal samples and 10 tumor adjacent normal samples), then, bioinformatic technique was used to construct the miRNA-mRNA interaction pathways of these miRNA using the differentially expressed miRNA.Results: A total of 194 differentially expressed miRNA were obtained, and the screened miRNA were involved in extensive post-transcriptional regulations of gene expression such as MAPK,WNT,TGF-β signaling pathway.Conclusion: The differentially expressed miRNA in colorectal cancer tissue may indicate that the miRNA with different functions or from
