摘要:
目的:观察两种方式注射白消安对SD大鼠生精功能的影响,探讨以其建立SD大鼠生精功能障碍模型的可行性。方法:观察腹腔注射白消安(分别给予15、20、30、40 mg/kg白消安)与睾丸内注射白消安(分别给予0.1、0.2、0.3和0.4 mg/kg白消安)对SD大鼠存活率、血常规以及睾丸病理Johnsen评分的影响,均设空白对照组、溶剂对照组和阴性对照组。结果:各腹腔注射组大鼠均在30 d内全部死亡,死亡前均表现为虹膜变灰色、眼睑结痂、活动迟缓,另有部分大鼠出现鼻周肛周血迹,血常规检测发现大鼠红细胞、血红蛋白和血小板较空白对照组降低(P<0.05),睾丸病理Johnsen评分与空白对照组比较,差异无统计学意义(P>0.05)。各睾丸内注射组大鼠观察30 d内无死亡,血常规检测结果显示大鼠红细胞、血红蛋白及血小板与正常对照组比较,差异无统计学意义(P>0.05),睾丸病理Johnsen评分均低于正常对照组(P<0.05)。结论:腹腔注射白消安,SD大鼠死亡率极高,而睾丸内注射白消安未发现对大鼠有明显毒害作用且0.1、0.2、0.3及0.4 mg/kg的剂量均可建立SD大鼠生精功能障碍模型。
Abstract:
Aim: To observe the effect of two methods of injecting busulfan to SD rats, and analyze the feasibility of establishing SD rat model of spermatogenesis dysfunction.Methods: Intraperitoneal injection of busulfan(15,20,30 and 40 mg/kg)and testicular injection of busulfan(0.1,0.2,0.3,and 0.4 mg/kg)were performed in the experiment, and survival rate, routine blood test and testicular pathology analysis were compared between them. The rats not treated(blank control)or given solvent or double distilled water were used as control groups.Results: Ⅰ)SD rats that were given busulfan by intraperitoneal injection all died within 30 d; before death the rats showed dimmed iris, eyelid crusting, slow activity and so on, and some rats showed bleeding around nose and anus. Blood analysis showed that red blood cells, hemoglobin and platelets of the groups given busulfan at different doses decreased significantly compared with blank control group(P<0.05). Johnsen score had no significant difference between the groups given busulfan at different doses and blank control group(P>0.05). Ⅱ)SD rats that were given busulfan by testicular injection were all alive within 30 days. Blood analysis revealed that red blood cells, hemoglobin and platelets of the groups given busulfan at different doses had no significant difference from those of blank control group(P>0.05),while Johnsen score of the groups given busulfan at different doses was significantly lower than that of blank control group(P<0.05).Conclusion: Intraperitoneal injection of busulfan in rats results in a high mortality. Testicular injection of busulfan(0.1~0.4 mg/kg)in rats showed no significant toxic effects and could be used to establish the model of spermatogenesis dysfunction.
