摘要:
目的:建立黄曲霉菌感染小鼠角膜动物模型,并观察病变后不同时期角膜组织病理学变化。方法:用培养接种棒对40只C57BL/6小鼠进行仿真临床角膜划痕导致真菌感染。在第1、3、5、7和14天行裂隙灯显微镜检查评分,随后处死小鼠制作角膜铺片,免疫荧光染色后进行激光扫描共聚焦显微镜观察。结果:术后第1、3和5天,接种眼临床评分升高迅速,在第5天达到峰值,随后第7和14天临床评分逐渐降低;术后第1天接种眼角膜缘组织已有中性粒细胞浸润、角膜巩膜缘血管扩张充血等明显急性炎症变化,第3天角膜巩膜缘出现新生血管芽,中性粒细胞向角膜中央溃疡灶浸润、聚集,第5天中性粒细胞浸润进一步加重,见侵入角膜基质层放射状生长的新生血管,第7天中性粒细胞浸润仍较严重,新生血管向角膜中央溃疡灶生长明显,管腔增粗,第14天中性粒细胞明显减少,新生血管萎缩、减少。结论:成功建立黄曲霉菌角膜炎小鼠动物模型;感染第5天可能是角膜炎症过程中的关键时间窗。
Abstract:
Aim: To establish an effective mouse model of simulated clinical corneal fungal infections and observe the pathological changes of corneal tissue lesions after different periods of fungal keratitis.Methods: Forty C57BL/6 mice models were inoculated with a culture stick simulated clinical corneal scratches which caused fungal infections.On the 1st,3rd, 5th,7th and 14th day, infectious corneas were observed and given clinical scoring by slitlamp biomicroscope,and then the mice were killed to make corneal stretched preparation. After immunofluorescence staining, the corneal tissue were observed with laser scanning confocal microscope. Results: The clinical scoring increased sharply on the 1st,3rd and 5th day after inoculation and peaked on the 5th day, then decreased on the 7th and 14th days. On the 1st day,the eye limbal tissue had neutrophil infiltration,corneal limbal vascular dilatation and congestion and other significant acute inflammatory changes.On the 3rd day,there were limbal corneal neovascularization buds, and neutrophil infiltration to gather corneal ulcer lesions. On the 5th day neutrophil infiltration further aggravated and invasive corneal stroma radial growth of new blood vessels could be seen. On the 7th day neutrophil infiltration was still severe, neovascularization obvious grew to the central cornea ulcer lesions,and luminal enlargement could be found on the 14th day neutropenia decreased significantly, and neovascularization atrophy reduced. Conclusion: An animal model of aspergillus keratitis has been successfully established. The key time window is the 5th day during the inflammation process of fungal keratitis.
