摘要:
目的:建立一种新的小鼠灌胃器,评价其灌胃效果。方法:24只清洁级昆明小鼠随机分为两组,分别采用常规小鼠灌胃针(常规组)和新建立的小鼠灌胃器(新型灌胃器组)灌胃,每只小鼠按照10 μL/g灌注用生理盐水配制的含体积分数50%的蓝墨水,记录两组灌胃操作时间(从固定好小鼠到灌胃结束)和成功率(解剖发现胃部有蓝色液体为灌胃成功标志),比较两组的灌胃效果。结果:常规组操作时间为(34.39±7.58) s,新型灌胃器组操作时间为(14.23±1.43) s,新型灌胃器组操作时间短于常规组(P<0.001)。新型灌胃器组无一只因操作不当引起小鼠食道损伤或误灌入气道死亡,药物顺利到达胃部,成功率达100.0%;常规组成功率为83.3%,有2只因灌胃针穿透食管使药物进入胸腔。结论:制作的新型小鼠灌胃器灌胃操作简单、无创,有助于提高动物福利和减少科学的变异性,值得推广使用。
Abstract:
Aim: To establish a novel oral gavage device for mice and evaluate its effect. Methods: A total of 24 clean grade KM mice were randomly allocated into two groups and administered saline configurated with 50% volume fraction of blue ink(10 μL/g) using normal oral gavage device or novel oral gavage device,respectively. Moreover, the operation time(from mice being fixed in hand to the end of oral gavage) and success(blue liquid could be found in the stomach) rate were recorded.Results: The operation time of normal oral gavage device group was (34.39±7.58) s, which was longer than that of the novel oral gavage device group[(14.23±1.43) s](P<0.001). There was none case with esophagus or airway injury induced by improper operation when using the novel oral gavage device,and the success rate was 100.0%. However, two mice were penetrated through esophagus by gavage needle when using normal oral gavage device,and the success rate was 83.3%. Conclusion: The novel oral gavage device is simple, noninvasive and disposable in improving animal welfare and decrease scientific variability.
