摘要:
目的:探讨瑞舒伐他汀对心肌肥厚大鼠心肌组织中自噬相关蛋白Beclin-1表达的影响。方法:健康雄性SD大鼠75只,随机分为假手术组、模型组和RSV组,每组25只,模型组和RSV组通过腹主动脉缩窄术建立心肌肥厚模型,术后分别给予生理盐水和瑞舒伐他汀(4 mg/kg)灌胃,1次/d,连用4周。4周后测定心脏指数,用RT-PCR和Western blot法分别检测大鼠左心室心肌组织中Beclin-1 mRNA和蛋白的表达水平。结果:与假手术组比较,模型组大鼠心肌明显肥厚,心脏指数升高,心肌组织中Beclin-1 mRNA和蛋白表达上调(P<0.05); 与模型组比较,RSV组大鼠心肌无明显肥厚,心脏指数降低,心肌组织中Beclin-1 mRNA和蛋白表达下降(P<0.05)。结论:瑞舒伐他汀可抑制压力超负荷性心肌肥厚,其作用机制可能与降低自噬水平有关。
Abstract:
Aim: To investigate the effect of rosuvastatin(RSV)treatment on Beclin-1 expression in myocardial tissue of rats with myocardial hypertrophy due to autophagy.Methods: A total of 75 healthy male SD rats were randomly allocated into 3 groups:sham operation group, model group, and RSV group, 25 rats in each group. Model group and RSV group were given normal saline and RSV(4 mg/kg)by gavage,respectively, one time a day and for 4 weeks. The heart samples were collected after four weeks to detect the expressions of Beclin-1 mRNA and protein using RT-PCR and Western blot.Results: Compared with sham operation group, myocardial hypertrophy, cardiac index and the expressions of Beclin-1 mRNA and protein in the model group increased(P<0.05). Compared with the model group, there was no obvious cardiac hypertrophy,and cardiac index,the expressions of Beclin-1 mRNA and protein were lower in RSV group(P<0.05).Conclusion: RSV could inhibit pressure overload cardiac hypertrophy, and the mechanism may be related with the decreased expression of Beclin-1.
