摘要:
目的:观察低能激光照射(LLLI)对大鼠心肌梗死区及其边缘组织中miRNA(简称miR)-21表达的影响。方法:将110只SD大鼠分为假手术组(30只)、对照组(40只)和治疗组(40只)。对照组和治疗组通过结扎左冠状动脉前降支制作心肌梗死模型,假手术组同部位只穿线不结扎,3周后治疗组采用LLLI处理梗死区域,对照组和假手术组照射同一区域但治疗仪不接通电源,各组分别于照射后1 h、24 h、48 h、72 h、7 d取梗死及边缘组织,采用RT-PCR法检测miR-21的表达。结果:梗死中心和边缘区比较,不同组别之间miR-21表达量比较差异有统计学意义(P<0.001),处理后不同时间miR-21表达量比较差异有统计学意义(P<0.001); 治疗组梗死区miR-21的表达在LLLI处理后各时间点的表达量均高于对照组(P<0.05); 治疗组梗死边缘miR-21的表达量在48 h达到峰值,且不同时间点的表达量均高于对照组和假手术组(P<0.05)。结论:LLLI处理梗死心肌能有效上调梗死及边缘区域miR-21的表达,有较好的心肌保护作用。
Abstract:
Aim: To observe the effects of low-level laser irradiation(LLLI)on miRNA(miR)-21 expression in myocardial infarct area and the edge tissue.Methods: A total of 110 SD rats were randomly allocated into sham operation group(30 rats), control group(40 rats), and the treatment group(40 rats).Myocardial infarction model in control and treatment groups was made by ligation of the left anterior descending coronary artery.After three weeks the treatment group was given LLLI, the control group and the sham operation group was irradiated at the same site but the device did not power on.At 1 h, 24 h, 48 h, 72 h, 7 d after irradiation, miR-21 expression was detected using RT-PCR.Results: miR-21 expression was significantly different between different groups and between the center and edge of the infarct area(P<0.001), miR-21 expression at different time points after treatment also had significant difference(P<0.001). The expression of miR-21 in the treatment group was higher than that in the control group at each time point after LLLI treatment(P<0.05), miR-21 expression level peaked at 48 h at the edge of the infarct area in treatment group,and the expression level at different time points was higher than control group and the sham operation group(P<0.05).Conclusion: LLLI treatment could increase the miR-21 expression in core and edge of infarct area, and has better myocardial protection function.
