摘要:
目的:观察虎杖苷对白血病细胞株K562细胞增殖、凋亡的影响,并探讨其可能机制。方法:以不同浓度(0、5、10、20、40、80、160、320 μmol/L)虎杖苷处理K562细胞12、24、48 h后,采用CCK-8法检测细胞增殖抑制率。另取对数生长期的K562细胞,分为3组,观察组加入80 μmol/L的虎杖苷,阴性对照组加入DMSO,空白对照组不处理,培养24 h后,采用Annexin V-FITC/PI双染法检测细胞凋亡,流式细胞仪检测细胞周期,Western blot检测p-Akt、p-mTOR、p70S6K、Bcl-2、Bax、Caspase-3蛋白表达水平。结果:虎杖苷能有效抑制K562细胞增殖,呈时间和剂量依赖性(P<0.001)。80 μmol/L虎杖苷作用24 h后,K562细胞凋亡率增加(P<0.001); 细胞周期出现G0/G1期至S期的阻滞(P<0.001); p-Akt、p-mTOR、p70S6K、Bcl-2蛋白表达水平下降(P<0.05),Bax、Caspase-3蛋白表达水平升高(P<0.05)。结论:虎杖苷可有效抑制K562细胞增殖并诱导其凋亡,可能是通过抑制Akt/mTOR/p70S6K信号通路及调控相关凋亡蛋白的表达来实现的。
Abstract:
Aim: To investigate the effect of polydatin on the cell proliferation and apoptosis of leukemia K562 cell line and to study its related molecular mechanism.Methods: Ⅰ:After treatment with different concentrations(0,5,10,20,40,80,160,320 μmol/L)of polydatin for 12, 24, 48 h, the proliferation rate of K562 cells was tested by CCK-8 assay. Ⅱ:K562 cells were allocated into 3 groups,the observation group and negative control group were treated with 80 μmol/L polydatin and DMSO,respectively,and blank control group was not treated.After 24 h, the apoptosis and cell cycle of K562 cells were detected by Annexin V-FITC/PI staining and flow cytometry, respectively. The protein expressions of p-Akt,p-mTOR,p70S6K,Bcl-2,Bax and Caspase-3 were detected by Western blot.Results: The proliferation rate of K562 cells was inhibited by polydatin in a time- and dose-dependent manner(P<0.001). After 24 h treatment with 80 μmol/L polydatin, the apoptosis rate of K562 cells increased(P<0.001),and there was an increase in G0/G1 phase cells and a decrease in S phase cells(P<0.001); polydatin could down-regulate the expressions of p-Akt,p-mTOR,p70S6K,and Bcl-2 and up-regulate the expressions of Bax and Caspase-3(P<0.05).Conclusion: Polydatin could effectively inhibit proliferation and induce apoptosis of K562 cells, which may be via inhibiting the Akt/mTOR/p70S6K signaling pathway and regulating the expressions of apoptosis related proteins.
