摘要:
目的:研究不同饮水氟暴露水平人群中雌激素受体(ER)α基因PvuⅡ多态性与血清钙、镁含量的关系。方法:采用整群抽样的方法,在高氟区、改水区和对照区选择当地出生或居住5 a以上18~55岁人群为观察对象,采用PCR-RFLP法检测ERα PvuⅡ多态性,采用原子吸收分光光度法测定血清钙、镁含量,采用氟离子选择电极法测定尿氟含量。结果:改水组男性血清钙、镁含量和女性血清镁含量均低于高氟组和对照组(P<0.05); 女性高氟组血清钙含量高于对照组和改水组(P<0.05)。多重线性回归结果显示,尿氟水平是人群血清钙的影响因素(β=0.139,P=0.003),尿氟与ERα PvuⅡ PP基因型对血清镁含量的影响存在交互作用(β=-0.046,P=0.042)。结论:长期氟暴露可影响人群血清钙、镁含量; ERα基因PvuⅡ多态性可能影响氟暴露地区人群血清镁含量。
Abstract:
Aim: To study the relationship between estrogen receptor α(ERα)gene PvuⅡ polymorphism and serum calcium and magnesium levels in adults with different fluoride exposure in drinking water.Methods: Local adults who were born there or lived for at least 5 years and aged 18-55 years were recruited as the subjects by cluster sampling in the high fluoride villages, the defluoridation project villages and the control villages. PCR-RFLP was used to determine ERα PvuⅡ genotypes. Serum calcium and magnesium levels were determined by atomic absorption spectrometry method. The fluoride concentration in urine was determined by fluoride ion selective electrode method.Results: Serum calcium and magnesium levels in males and serum magnesium level in females from the defluoridation project group were lower than those from the high fluoride group and the control group, respectively(P<0.05). Serum calcium level in females from the high fluoride group was higher than those from the defluoridation project group and the control group(P<0.05). Multiple linear regression analysis showed that urine fluoride was the major factor affecting serum calcium level(β=0.139,P=0.003), and the interaction between urine fluoride and ERα PvuⅡ PP genotype on serum magnesium level was significant(β=-0.046,P=0.042).Conclusion: Serum calcium and magnesium levels would be affected by long-term fluoride exposure. There was an association between ERα PvuⅡ polymorphism and serum magnesium level in adults exposed to fluoride in drinking water.
