摘要:
目的:检测游离二氧化硅所致肺成纤维细胞转分化过程中可能存在的DNA甲基化差异基因并探讨其可能作用机制。方法:原代培养肺成纤维细胞和肺泡巨噬细胞,建立共培养的矽肺体外模型,采用甲基化DNA免疫共沉淀测序技术检测分析不同染毒时间(0、24、48 h)的肺成纤维细胞之间的DNA甲基化差异基因,并采用DAVID工具对检测到的甲基化差异基因进行KEGG的通路富集分析。结果:共发现8 791个基因的DNA甲基化程度在3个染毒时间组中均存在差异,DAVID工具分析发现这些差异主要集中在代谢、环境信息过程、细胞过程、生物体系统、人类疾病相关通路等通路中。结论:矽肺形成过程中存在大量DNA甲基化程度发生改变的基因,且这些基因涉及多个通路。
Abstract:
Aim:To examine the different DNA methylated genes and possible mechanism in the transdifferentiation of lung fibroblasts caused by free SiO2.Methods: The primary macrophages and fibroblasts of SD rats were cultured, and then co-cultured. DNA methylation in fibroblasts with different infected time(0,24,48 h)was detected by MeDIP-seq.Results: A total of 8 791 different DNA methylated genes were discovered in all the three groups, and a further more analysis about those genes in KEGG pathway was performed by DAVID. The different DNA methylated genes were found enriched in metabolism, environmental information processing, cellular processes, organismal systems, human diseases pathway,etc.Conclusion: A large number of different DNA methylated genes and pathways exist in the pathogenesis of silicosis, which is valuable for further study.
