摘要:
目的:观察环丙沙星(CIP)和左氧氟沙星(LVFX)对豚鼠右心室乳头肌动作电位的影响。方法:取豚鼠右心室乳头肌,用氧饱和的台式液持续表面灌流。采用累积浓度给药法于灌流液中加入CIP或LVFX,使药物终浓度依次达到0、1、10和100 μmol/L。用标准微电极技术记录乳头肌加药前后静息电位(RP)、动作电位幅值(APA)、动作电位复极至50%的时程(APD50)及APD90。以司帕沙星(SPX)为阳性对照药。每组6个样本。结果:3个药物对RP、APA均没有影响(P>0.05)。SPX可延长APD50 和APD90,并且作用随浓度增加而增强(P<0.001)。LVFX不延长APD50(P=0.110),可延长APD90,并且该作用随浓度增加而增强(P<0.001)。CIP 对APD50无影响,仅在100 μmol/L时表现出延长APD90的作用(P=0.002)。结论:大剂量CIP和LVFX可引起豚鼠右心室乳头肌动作电位时程延长,具有潜在的心脏毒性。
Abstract:
Aim: To examine the effects of ciprofloxacin(CIP)and levofloxacin(LVFX)on cardiac action potentials in right ventricle papillary muscles of guinea pigs.Methods: The right ventricle papillary muscles of guinea pigs were treated with oxygen-saturated Tyrode continuous perfusion. Cumulative concentration dosing method was used to join CIP or LVFX in perfusion and the drug concentration in turn reached to 0,1,10 and 100 μmol/L. The time of resting potential(RP),amplitude of action potential(APA),action potential duration of 50% repolarization(APD50)and APD90 were recorded by conventional microelectrode techniques. Sparfloxacin(SPX)was the positive control, and 6 pigs in each group.Results: The three drugs had no effect on RP or APA(P>0.05). SPX could prolong APD50 and APD90, and the effects enhanced with the concentration increasing(P<0.001). LVFX could not prolong APD50(P=0.110), but could prolong APD90, and this effect enhanced with the increase of the concentration(P<0.001). CIP had no effect on APD50,and could extend APD90 only at dose of 100 μmol/L(P=0.002).Conclusion: CIP and LVFX at large dose could prolong the action potential duration of right ventricle papillary muscles of guinea pigs, and have potential cardiac toxicity.
