摘要:
目的:探讨腺苷A2A受体对新生小鼠离体延髓脑片基本节律性呼吸放电(RRDA)的调节作用。方法:制备新生小鼠离体延髓脑片,分别使用含0.0、2.5、5.0、10.0、20.0 nmol/L腺苷A2A受体激动剂CGS21680或拮抗剂SCH58261的人工脑脊液(ACSF)灌流脑片,每个浓度灌流10 min,在每个浓度灌流第6 min时,使用吸附电极记录脑片RRDA,分析吸气时程(TI)、放电积分幅度(IA)和呼吸周期(RC)。结果:CGS21680可以缩短RC,增加TI和IA(P<0.001),对RRDA有兴奋作用。SCH58261则可以延长RC,减小TI和IA(P<0.001),对RRDA有抑制作用。结论:腺苷A2A受体参与调节新生小鼠延髓RRDA,激活腺苷A2A受体对RRDA有兴奋作用。
Abstract:
Aim: To investigate the effect of adenosine A2A receptor on basic rhythmic respiratory discharge activity(RRDA)of medulla oblongata slice from neonatal mice.Methods: Medulla oblongata slices in vitro from neonatal mice were prepared, and then perfused with artificial cerebrospinal fluid(ACSF)which containing 0.0, 2.5, 5.0, 10.0, 20.0 nmol/L adenosine A2A receptor agonist CGS21680 or antagonist SCH58261 for 10 min. At the 6th min of perfusion, adsorption electrode was used to record the inhale duration(TI), discharge integral amplitude(IA)and respiratory cycle(RC).Results: CGS21680 could shorten RC, increase TI and IA(P<0.001), excite RRDA; SCH58261 could extend RC, reduce TI and IA(P<0.001),and has inhibitory effect on RRDA.Conclusion: The adenosine A2A receptor may play an excitatory role in RRDA of medulla oblongata in neonatal mice.
