摘要:
目的:检测重度子痫前期患者胎盘组织中NFAT5、MCP-1的表达并探讨二者与新生儿结局的关系。方法:采用免疫组化法检测30例早发型重度子痫前期(早发型组)、30例晚发型重度子痫前期(晚发型组)及30例足月妊娠(对照组)孕妇胎盘组织中NFAT5、MCP-1蛋白的表达情况,采用RT-PCR法检测胎盘组织中NFAT5、MCP-1 mRNA的表达情况,分析早发型组2种蛋白的相关性及与新生儿结局的关系。结果:3组胎盘组织中均有NFAT5和MCP-1蛋白的表达; 早发型组2种因子的蛋白及mRNA表达水平均高于晚发型组(P<0.05); 晚发型组与对照组比较,差异无统计学意义(P>0.05)。早发型组胎盘组织中NFAT5与MCP-1蛋白的表达呈正相关(r=0.779,P<0.001); 而NFAT5和MCP-1蛋白分别与新生儿体重(r=-0.760、-0.560,P<0.05)、胎盘质量(r=-0.583、-0.464,P<0.05)呈负相关。结论:早发型子痫前期胎盘组织中NFAT5、MCP-1的表达均高于晚发型子痫前期,说明两者可能有不同的发病机制。
Abstract:
Aim: To investigate the expressions of NFAT5 and MCP-1 and their relationship with the perinatal outcome in patients with severe preeclampsia.Methods: The expressions of NFAT5 and MCP-1 in 30 case of early-onset preeclampsia(EOPE group), 30 cases of late-onset preeclampsia(LOPE group)and 30 normal control(control group)tissue were detected by immunohistochemistry. RT-PCR was conducted to analyze the expressions of NFAT5 and MCP-1 mRNA in placental tissue. The relationship among the expressions of NFAT5 and MCP-1 protein and the perinatal outcome in EOPE group was analyzed.Results: NFAT5 and MCP-1 protein and mRNA expressions in the EOPE group were both higher than those in the LOPE group(P<0.05). However, there was no difference between the LOPE group and the control group(P>0.05).The expression of NFAT5 was positively correlated with the expression of MCP-1 in the EOPE group(r=0.779,P<0.001). NFAT5 and MCP-1 expressions in the EOPE group was negatively correlated with neonatal weight(r=-0.760,-0.560,P<0.05)and the placental weight(r=-0.583,-0.464,P<0.05).Conclusion: The expressions of NFAT5 and MCP-1 in the EOPE group are both higher than those in the LOPE group, which suggests that they may have different pathogenesis.
