摘要:
目的:构建FOXQ1慢病毒载体,探究FOXQ1过表达对人脐带间充质干细胞(hUC-MSCs)衰老的影响。方法:PCR扩增FOXQ1序列,构建慢病毒pHBLV-FOXQ1-Puro重组载体,包装病毒、感染hUC-MSCs(过表达组),感染空载体病毒的hUC-MSCs为对照,分别采用实时荧光定量PCR、Western blot检测2组细胞FOXQ1 mRNA及蛋白的表达水平,采用CCK-8法检测细胞增殖能力的变化,采用SA-β-Gal与AP染色检测hUC-MSCs衰老表型。结果:慢病毒pHBLV-FOXQ1-Puro重组载体构建成功,病毒包装后感染hUC-MSCs效果良好。过表达组FOXQ1蛋白及mRNA表达水平均高于对照组(P<0.05)。过表达组细胞增殖能力较强,FOXQ1过表达可以延缓hUC-MSCs衰老。结论:成功建立FOXQ1稳定过表达的hUC-MSCs细胞系,FOXQ1过表达可以延缓hUC-MSCs衰老。
Abstract:
Aim: To construct the FOXQ1 overexpression lentivirus vector and explore the effect of FOXQ1 overexpression on human umbilical cord mesenchymal stem cells(hUC-MSCs)senescence.Methods: After amplification of FOXQ1 sequences, the recombinant lentivirus vector(pHBLV-FOXQ1-Puro)was constructed. The hUC-MSCs overexpressing FOXQ1 infected by the prepared vector and screened by puromycin screening. The hUC-MSCs infected by the empty vector were used as control. The protein and mRNA expression levels of FOXQ1 were measured by Western blot and qPCR. CCK-8 assay was used to test the ability of cell proliferation. SA-β-Gal and AP staining assay were used to detect the effect of FOXQ1 overexpression on cell senescence.Results: pHBLV-FOXQ1-Puro recombinant vector was constructed successfully, and its infection was effective. The protein and mRNA expression levels of FOXQ1 overexpression group were significantly higher than control group(P<0.05). CCK-8 results showed that FOXQ1 overexpression could enhance the ability of cell proliferation. SA-β-Gal and AP staining results showed that FOXQ1 overexpression could delay senescence of hUC-MSCs.Conclusion: The hUC-MSCs overexpressing FOXQ1 have been successfully constructed, and FOXQ1 overexpression could delay the senescence of hUC-MSCs.
