摘要:
目的:探讨上调FOXC1的表达对卵巢浆液性腺癌SKOV3细胞增殖及侵袭的影响。方法:分别以重组FOXC1慢病毒、慢病毒空载体稳定感染SKOV3细胞(实验组和载体对照组),并以未感染慢病毒的SKOV3细胞为空白对照组,分别应用qRT-PCR和Western blot法检测3组细胞中FOXC1 mRNA和蛋白的表达情况,应用CCK-8法检测3组细胞接种24、48、72 h后的增殖能力,用Transwell小室检测接种24 h细胞的体外侵袭能力。结果:实验组SKOV3细胞FOXC1 mRNA及蛋白的表达均较载体对照组及空白对照组升高(P<0.05)。与空白对照组及载体对照组相比,实验组SKOV3细胞感染不同时间的增殖活性均降低,侵袭能力亦降低(P<0.05)。结论:上调FOXC1的表达能够抑制SKOV3细胞的增殖及侵袭能力。
Abstract:
Aim: To explore the changes of proliferation and invasion in SKOV3 cells after up-regulating the expression of FOXC1.Methods: SKOV3 cells were divided into 3 groups: cells in experimental group were infected with p-EGFP-lentiviral vector with overexpressed FOXC1 plasmids, cells in vector control group were infected with empty p-EGFP-lentiviral vector, and cells in blank control group, not infected. The expressions of FOXC1 mRNA and protein were detected by qRT-PCR and Western blot,respectively. The cell proliferation after 24,48 and 72 h infection and cell invasion after 24 h infection were observed by CCK-8 kit and Transwell invasion assay,respectively.Results: The expressions of FOXC1 mRNA and protein in SKOV3 cells in experimental group were significantly higher than those in the vector control and blank control groups(P<0.05).Compared with blank control group and vector control group, the proliferation activity of SKOV3 cells in experimental group was reduced at different time points, so was the invasive ability(P<0.05).Conclusion: Up-regulation of FOXC1 may suppress cell proliferation and invasion of SKOV3 cells.
