摘要:
目的:基于荧光碳量子点(CQDs)的催化特性,建立CQDs -鲁米诺化学发光新体系用于2-甲氧基雌二醇(2-ME)的含量测定。方法:利用微波热解法合成荧光CQDs,采用透射电镜及光谱技术对其进行表征。将CQDs应用于鲁米诺发光体系中,考察不同浓度试剂对发光强度的影响,在最优条件下,对冻干粉剂及血浆样品中的2-ME含量进行测定。结果:CQDs溶液可以增强鲁米诺-铁氰化钾体系的发光信号,因2-ME对这一体系的发光有抑制作用,且该作用与2-ME的浓度呈正比关系,因此建立了检出限为5.4×10-10 g/mL,RSD为1.8%(n=11)的2-ME含量测定的新方法。结论:该方法灵敏度高、简单快速,适用于药物制剂及生物样品中2-ME的含量测定。
Abstract:
Aim: To establish a new carbon quantum dots(CQDs)-luminol chemiluminescence system for the determination of anticancer drug 2-methoxyestradiol(2-ME)based on the catalysis properties of CQDs.Methods: Fluorescent CQDs was synthesized by microwave pyrolysis and characterized by TEM and spectroscopy. The influence of luminous reagents concentration on intensity were investigated and the reaction mechanism was explored in this paper. Under the optimal conditions, the 2-ME content in freeze-dried powder and biological samples were tested.Results: This CQDs can enhance the fluorescent intensity of luminol-potassium ferricyanide system. It was found that 2-ME had significant inhibition on fluorescent intensity of the new system with concentration dependent manner. Based on this effect, the new determination method of 2-ME was developed with detection limit of 5.4×10-10 g/mL and the RSD(n=11)of 1.8% for 2-ME, respectively.Conclusion: This method is simple, accurate and rapid for the quantitative analysis of 2-ME in pharmaceutical preparations and biological samples with satisfactory results.
