摘要:
目的:探讨2,3,7,8-四氯二苯并二噁英(TCDD)对大鼠睾丸间质细胞凋亡相关蛋白表达及ROS 含量的影响。方法:用不同剂量TCDD(0、1、5、10、50 nmol/L)处理大鼠睾丸间质细胞24 h后,采用MTT法检测细胞增殖抑制率,利用荧光探针DCFH-DA检测细胞ROS含量,Annexin V-FITC/PI染色联合激光共聚焦显微镜观察细胞凋亡情况,采用Western blot法检测细胞中凋亡相关蛋白Bax、 Bcl-2、 Caspase-3的表达水平。结果:随着TCDD剂量的增加,细胞增殖抑制率和ROS含量逐渐升高(P<0.05),细胞中Bax、Caspase-3蛋白相对表达量逐渐增加,而Bcl-2蛋白相对表达量逐渐降低(P<0.05)。结论:TCDD可诱导大鼠睾丸间质细胞凋亡。
Abstract:
Aim: To explore the effects of TCDD on expressions of apoptosis related proteins and ROS content in leydig cells of rats.Methods: Leydig cells from rats were treated with different doses of TCDD(0,1,5,10,50 nmol/L)for 24 hours, and the proliferation inhibition rate of leydig cells was determined by MTT assay, ROS content in leydig cells was determined by DCFH-DA method, apoptosis was observed by confocal microscopy combined Annexin V-FITC/PI staining, and the expressions of Bax, Caspase-3 and Bcl-2 proteins were detected by Western blot.Results: With the increase of the dose of TCDD, the proliferation inhibition rate and ROS content showed a rising trend(P<0.05), the expressions of Bax and Caspase-3 increased whereas that of Bcl-2 protein decreased(P<0.05).Conclusion: TCDD could significantly induce apoptosis of rat leydig cells.
