摘要:
目的:研究单侧Ⅱ类牵引对成年大鼠牵引侧颞下颌关节滑膜的影响。方法:180只8周龄的SD雄性大鼠采用随机数字表法分为3组:A组(实验组),在右下第一磨牙和右上切牙之间固定Ni-Ti拉簧,加力20 g,建立单侧Ⅱ类牵引模型; B组(假手术组),同A组固定拉簧,不加力; C组(空白组),不固定拉簧。每组按时间点(第1、3、7、14、21、28天)处死大鼠,取右侧关节双板区的滑膜组织进行HE染色和IL-1β、VEGF免疫组化染色,观察滑膜的组织学变化,检测IL-1β、VEGF的表达,并分析A组滑膜的病理评分和IL-1β、VEGF表达量之间的关系。结果:A组各时间点IL-1β和VEGF的表达均比相应时间点B组、C组高(P<0.001)。相应时间点A组的病理评分与IL-1β、VEGF的表达呈正相关(r=0.912、0.930,P均<0.001)。结论:单侧Ⅱ类牵引可引起颞下颌关节滑膜发生组织学改变,这种改变与滑膜组织IL-1β、VEGF的表达有关。
Abstract:
Aim: To study the effects of unilateral class Ⅱ traction on temporomandibular joint(TMJ)synovial membrane in adult rats.Methods: A total of 180 8-week-old male SD rats were randomly allocated into three groups:group A(experimental group),fixed Ni-Ti spring loaded about 20 grams between the first molar and the upper incisor of right; group B(sham operation group),fixed Ni-Ti spring but not loaded; group C(control group), fixed no spring. Rats of each group were sacrificed at the time points(the 1st,3rd,7th,14th,21st,28th day), then the right synovial tissue of double plate area was prepared,and HE staining and immunohistochemical staining were used to observe the histological changes of synovial tissue and detect the expressions of IL-1β and VEGF, respectively. The relationship between the pathological score and the expressions of IL-1β and VEGF in group A was analyzed.Results: The expressions of IL-1β and VEGF in group A at each time point were higher than those of group B and group C(P<0.001).There was a positive correlation between the pathological score and the expression of IL-1β or VEGF in group A(r=0.912, 0.930, P<0.001).Conclusion: Unilateral class Ⅱ traction could cause histological changes of TMJ synovial tissue.There is a positive correlation between the histological changes and the expression of IL-1β or VEGF.
