摘要:
目的:探讨miRNA-138在TGF-β1诱导的乳腺癌MDA-MB-231细胞上皮间质转化(EMT)发生中的作用。方法:0.005 g/L TGF-β1刺激MDA-MB-231细胞48 h,以未刺激细胞作为空白对照,RT-PCR和Western blot检测MDA-MB-231细胞E-cadherin和vimentin mRNA、蛋白表达的变化,RT-PCR检测miRNA-138表达的变化。MDA-MB-231细胞转染100 nmol/L miRNA-138拟似物48 h,以转染100 nmol/L miRNA无关序列细胞、单纯转染脂质体细胞和空白细胞作为对照,RT-PCR检测miRNA-138表达的变化,RT-PCR和Western blot检测MDA-MB-231细胞E-cadherin和vimentin mRNA、蛋白表达的变化。0.005 g/L TGF-β1刺激同时转染100 nmol/L miRNA-138拟似物48 h,以0.005 g/L TGF-β1刺激同时转染100 nmol/L miRNA无关序列和空白细胞作为对照,RT-PCR和Western blot检测MDA-MB-231细胞E-cadherin和vimentin mRNA、蛋白表达的变化。结果:与空白对照组相比,0.005 g/L TGF-β1刺激MDA-MB-231细胞48 h可降低E-cadherin表达,提高vimentin表达,抑制miRNA-138表达(P<0.05)。与其他3组相比,转染100 nmol/L miRNA-138 拟似物 48 h组MDA-MB-231细胞E-cadherin表达升高,而vimentin表达降低(P<0.05)。与其他2组相比,miRNA-138表达增加可部分逆转TGF-β1刺激诱导的MDA-MB-231细胞EMT的发生。结论:miRNA-138参与TGF-β1诱导的乳腺癌MDA-MB-231细胞EMT的发生。
Abstract:
Aim: To explore the role of miRNA-138 in TGF-β1 induced epithelial-mesenchymal transformation(EMT)in breast cancer.Methods: Firstly, MDA-MB-231 cells were activated by 0.005 g/L TGF-β1 for 48 h, and vacant MDA-MB-231 cells were used as control. Then RT-PCR and Western blot were used to check the mRNA and protein expression of E-cadherin and vimentin, and RT-PCR was used to check the expression level of miRNA-138 in MDA-MB-231 cells in two groups. Secondly, 100 nmol/L miRNA-138 mimics were transfected into MDA-MB-231 cells for 48 h, MDA-MB-231 cells transfected with 100 nmol/L miRNA scramble, Lipofectamine 2000 or vacant MDA-MB-231 cells were used as controls. Then RT-PCR and Western blot were used to check the mRNA and protein expression of E-cadherin and vimentin, and RT-PCR was used to check the expression level of miRNA-138 in MDA-MB-231 cells in these four groups. Finally, MDA-MB-231 cells were activated by 0.005 g/L TGF-β1 and transfected with 100 nmol/L miRNA-138 mimics simultaneously for 48 h, MDA-MB-231 cells activated by 0.005 g/L TGF-β1 and transfected with miRNA scramble or vacant MDA-MB-231 cells were used as controls. Then RT-PCR and Western blot were used to check the mRNA and protein expression of E-cadherin and vimentin in MDA-MB-231 cells in these three groups.Results: 0.005 g/L TGF-β1 activation could induce the occurrence of EMT by inhibiting the expression of E-cadherin, at the same time, promoting the expression of vimentin(P<0.05); furthermore, the expression of miRNA-138 was inhibited by the activation of TGF-β1(P<0.05). Transfection of miRNA-138 mimics could suppress the expression of vimentin and increase the expression of E-cadherin(P<0.05). In addition, increasing expression level of miRNA-138 could restrain the occurrence of EMT induced by TGF-β1 to some extent.Conclusion: miRNA-138 participates in EMT induced by TGF-β1 in breast cancer cell line MDA-MB-231.
