摘要:
目的:采用超高效液相色谱-质谱联用技术,考察胃癌患者与健康志愿者血浆代谢谱的差异,并筛查胃癌的生物标志物,为胃癌发病机制的探究和胃癌的初步诊断提供依据。方法:运用超高效液相色谱-四级杆-静电场轨道阱高分辨质谱联用技术对30例胃癌患者及30名健康志愿者的血浆样本进行代谢组学分析,采用正交偏最小二乘判别分析法对数据进行模式识别,分析胃癌患者与健康志愿者间的代谢谱差异,并筛选出胃癌患者的差异代谢物。结果:胃癌患者血浆中脯氨酸、苯丙氨酸等8种代谢物浓度上升,亮氨酸、赖氨酸等5种代谢物浓度下降; 而精氨酸和脯氨酸代谢、苯丙氨酸代谢等代谢通路均发生明显重编程,提示胃癌患者体内形成了新的代谢调控网络。结论:代谢通路的显著变化可能与胃癌的发病机制有关,筛选出的差异代谢物有望作为胃癌患者的潜在生物标志物。
Abstract:
Aim: To analyze the metabolic profiles in the plasma samples of gastric cancer patients as well as healthy controls and further explore potential biomarkers related to gastric cancer.Methods: Ultra high performance liquid chromatography-Q exactive orbit trap high-resolution mass spectrometry was applied to process the plasma samples of 30 gastric cancer patients and 30 healthy volunteers. Orthogonal to partial squares discriminant analysis was performed to identify differences in metabolic profiles between the two groups and select potential biomarkers of gastric cancer.Results: The concentrations of 8 metabolic biomarkers such as in the plasma of patients with proline ans phenylalanine cancer were significantly increased, while the concentrations of 5 metabolic biomarkers such as leucine and lysine were significantly decreased, while the levels of arginine and proline metabolic pathways, phenylalanine metabolism and other pathways were significantly reprogrammed, suggesting that gastric cancer patients had a new metabolomic regulatory network.Conclusion: Significant changes in metabolic pathway may be related to the pathogenesis of gastric cancer. The screened differential metabolites are expected to serve as potential biomarkers for patients with gastric cancer.
