摘要:
目的:探讨过敏性鼻炎(AR)患者外周血嗜酸性粒细胞富集群中IL-18、IL-18BP和IL-18R的表达。方法:收集15例AR患者和15名体检健康者(正常对照组)外周静脉血,分别用大籽蒿花粉、尘螨或梧桐花粉粗提液刺激,流式细胞术检测嗜酸性粒细胞富集群中IL-18+、IL-18BP+和IL-18R+细胞比例及平均荧光强度(MFI)。结果:过敏原刺激前,AR患者嗜酸性粒细胞富集群中IL-18BP+和IL-18R+细胞比例分别是正常对照组的1.879倍和1.698倍(P<0.05)。梧桐花粉粗提液刺激后,AR患者IL-18+ 细胞比例升高约33%(P<0.05)。AR患者IL-18+细胞和IL-18BP+细胞MFI高度相关(rS=0.783,P<0.001)。正常对照组IL-18、IL-18BP和IL-18R的表达在过敏原刺激前后差异均无统计学意义(P>0.05)。结论:嗜酸性粒细胞源的IL-18、IL-18BP和IL-18R可能在AR中起重要作用,IL-18、IL-18BP和IL-18R可能是治疗AR的潜在靶点。
Abstract:
Aim: To investigate the expressions of IL-18, IL-18BP and IL-18R in blood eosinophil-enriched cell population from patients with allergic rhinitis(AR).Methods: Peripheral blood samples were collected from 15 AR patients and 15 healthy volunteers, then respectively stimulated by the extracts of Artemisia pollen, dust mites, or Platanus pollen, and proportions and mean fluorescent intensity(MFI)of IL-18+, IL-18BP+ and IL-18R+ cells in blood eosinophil-enriched cells were detected by flow cytometry.Results: Before stimulation,the proportions of IL-18BP+ and IL-18R+ cells of AR group were 1.8 and 1.7- fold of those of normal control(P<0.05). In AR group, Platanus pollen extracts induced approximately 33% increase in proportion of IL-18+ cells(P<0.05). In addition, MFI of IL-18+ cells and that of IL-18BP+ cells were positively correlated in AR group(rS=0.783, P<0.001). No statistical significance was detected in the expressions of IL-18, IL-18BP or IL-18R in normal controls after the stimulation of allergen(P>0.05).Conclusion: IL-18, IL-18BP and IL-18R from eosinophils are likely to play a key role in AR. IL-18, IL-18BP, and IL-18R might become the potential targets for AR treatment.
