摘要:
目的:探讨二氧化铈纳米颗粒(nanoceria)预处理对大鼠缺血再灌注(IR)心肌细胞中核因子E2相关因子2(Nrf2)信号通路表达的影响。方法:选取健康成年大鼠50只,随机均分为5组:假手术组、模型组、nanoceria预处理1~3组。模型组、nanoceria预处理1~3组制备心肌IR动物模型(心肌缺血45 min后再灌注120 min),nanoceria预处理1~3组分别于造模术前24 h经尾静脉注射1~10、10~25和50 nm粒径的nanoceria悬液(2 mL/kg)。再灌注结束后,荧光定量PCR法检测再灌注区域心肌组织中Nrf2、醌氧化还原酶1(NQO1)及血红素氧化酶1(HO-1)mRNA表达水平,Western blot法检测胞核内Nrf2蛋白表达水平。结果:模型组心肌细胞内NQO1、HO-1 mRNA表达水平及胞核内Nrf2蛋白表达水平较假手术组增加(P<0.05),Nrf2 mRNA表达量无明显变化(P>0.05)。与模型组比较,nanoceria预处理1~3组Nrf2、NQO1和HO-1 mRNA及胞核内Nrf2蛋白表达水平增加(P<0.05), nanoceria预处理2组变化最显著(P<0.05)。结论:nanoceria预处理可能通过激活Nrf2信号通路,发挥心肌保护作用。
Abstract:
Aim: To research the influence of pretreatment of nanoceria on the expression of nuclear factor related factor 2(Nrf2)in cardiomyocytes of rats with ischemia-reperfusion(IR)injury.Methods: Fifty adult healthy rats were randomly allocated into 5 groups,and 10 in each group: the sham operation group, model group, and 3 pretreatment groups. The rats in model group and 3 pretreatment groups were made myocardial IR model(ischemia for 45 min and reperfusion for 120 min). Pretreatment group 1,2,3 were injected through the tail vein with 1-10, 10-25, and 50 nm nanoceria suspension(2 mL/kg)24 h before the operation. After the reperfusion, the expressions of Nrf2, NQO1 and HO-1 mRNA and the expression of Nrf2 protein in the nucleus in IR cardiomyocytes were detected by qRT-PCR and Western blot.Results: The expressions of NQO1,HO-1 mRNA, and Nrf2 protein in the nuclei in the model group was significantly higher than those in the sham operation group(P<0.05), but the expression of Nrf2 mRNA had no significant change(P>0.05).Compared with those of the model group, the expressions of Nrf2,NQO1,HO-1 mRNA and Nrf2 protein in the nuclei in the 3 pretreatment groups increased(P<0.05), and the changes of the above indexes in pretreatment group 2 were the most significant(P<0.05).Conclusion: The pretreatment with nanoceria may activate the Nrf2 signaling pathway, so as to achieve myocardial protection.
