摘要:
目的:探讨人脑胶质瘤组织中高迁移率族蛋白B1(HMGB1)及Toll样受体4(TLR4)的表达及其与肿瘤病理的相关性。方法:选取于郑州大学第五附属医院神经外科行“胶质瘤切除术”的胶质瘤患者87例为试验组,同期于该院行“颅内减压术”的患者20例为对照组; 术中分别留取胶质瘤组织标本和颅内减压的脑组织标本,应用免疫组化法和Western blot法检测2组脑组织中HMGB1、TLR4的表达,结合患者临床因素及术后肿瘤病理,分析HMGB1、TLR4表达水平与胶质瘤病理的关系。结果:试验组患者胶质瘤组织中HMGB1、TLR4阳性表达率均高于对照组患者脑组织(P均<0.001); 试验组患者胶质瘤组织中HMGB1及其受体TLR4表达水平与胶质瘤病理级别有关(P均<0.05),而与患者性别、年龄、胶质瘤位置、胶质瘤病理类型无关(P均>0.05); 试验组患者胶质瘤组织中HMGB1表达水平与TLR4表达水平呈正相关(rp=0.515,P<0.001)。结论:HMGB1、TLR4可能与胶质瘤发病机制、肿瘤病理密切相关。
Abstract:
Aim: To detect the expressions of high mobility group box-1(HMGB1)and Toll like receptor-4(TLR4)in human brain glioma tissue and analyze the correlation of HMGB1 and TLR4 expressions in human brain glioma tissue and glioma pathology.Methods: A total of 87 patients who undertook "glioma resection" were selected as trial group, and 20 patients who undertook "intracranial decompression" were selected as control group. The tissue specimens of glioma and intracranial decompression were obtained during operation, and the expressions of HMGB1 and TLR4 were detected by immunohistochemistry and Western blot. The correlation between expression levels of HMGB1, TLR4 and glioma pathology was analyzed by combining the clinical factors and postoperative tumor pathology.Results: Positive expression rates of HMGB1 and TLR4 in glioma tissue of the trial group were both higher than those in normal brain tissue of control group(P<0.001). Expression levels of HMGB1 and TLR4 in glioma tissue of the trial group were correlated with pathological grade of glioma(P<0.05), but not correlated with gender, age, location of glioma, or pathological type of glioma(P>0.05). In the trial group, HMGB1 expression level was positively correlated with TLR4 expression level(rp =0.515,P<0.001).Conclusion: HMGB1, TLR4 may correlate with pathogenesis and pathology of human glioma.
