摘要:
目的:评价脂肪干细胞(ADSCs)对糖尿病型大鼠勃起功能障碍(ED)的治疗效果,探讨ADSCs治疗ED的机制。方法:108只雄性SD大鼠,其中20只为正常对照,余88只大鼠通过腹腔注射STZ建立糖尿病模型。糖尿病大鼠行阿扑吗啡实验,将实验阴性者分为ED模型组(19只大鼠)和ADSCs治疗组(20只大鼠),前者海绵体移植PBS缓冲液,后者海绵体移植ADSCs悬液。4周后,在10 V刺激下测定海绵体内压(ICP)和平均动脉压(MAP),计算ICP/MAP。采用免疫荧光染色方法检测大鼠阴茎海绵体中vWF、 α-SMA、nNOS蛋白的表达。结果:①ADSCs增殖能力较强且可被诱导分化为脂肪细胞和成骨细胞。②正常组、ED模型组和ADSCs组的ICP/MAP值分别为(0.690±0.096)、(0.250±0.096)和(0.440±0.130),差异有统计学意义(P<0.05),ADSCs组明显高于ED模型组。③ADSCs治疗组大鼠阴茎海绵体vWF、 α-SMA和nNOS蛋白的表达均高于ED模型组(P<0.05)。结论:ADSCs疗法能够提高大鼠的勃起功能,增加大鼠阴茎海绵体vWF、α-SMA和nNOS蛋白的表达。
Abstract:
Aim:To evaluate the role of adipose tissue-derived stem cell(ADSCs)in erectile dysfunction(ED)treatment and explore the mechanism.Methods:A total of 108 male SD rats were enrolled in the experiment,20 were randomly divided into normal control group,and the remaining 88 rats were injected with STZ to induce diabetic model. Rats with diabetic mellitus were tested with APO test, and those with no erection were randomly allocated into ED model group(n=19)and ADSCs treatment group(n=20); the former group was intracavernously injected with PBS and the later one with ADSCs suspension.After 4 weeks of intervention,ICP and MAP of the rats were measured and the therapeutic effect was evaluated. The expressions of vWF,α-SMA, and nNOS in corpus cavernosum were detected by immunofluorescence staining.Results:ADSCs had a strong proliferation ability and could be induced to differentiate into adipocytes and osteoblasts. The ICP/MAP of normal control group, ED model group and ADSCs treatment group was(0.690±0.096),(0.250±0.096)and(0.440±0.130), and the difference was significant(P<0.05). ICP/MAP in ADSCs treatment group was significantly higher than that in ED model group.In ADSCs treatment group, the expressions of vWF,α-SMA and nNOS were higher than those in the ED model group(P< 0.05).Conclusion: ADSCs can improve erectile function in rats.ADSCs can enhance the expressions of vWF,α-SMA and nNOS in corpus cavernosum of ED rats.
