摘要:
目的:研究Toll样受体4(TLR4)对肺炎链球菌诱导的A549细胞凋亡的影响。方法:用肺炎链球菌诱导A549细胞后感染TLR4 siRNA慢病毒,检测TLR4 siRNA对细胞中TLR4表达的沉默效果,流式细胞术检测A549细胞凋亡的变化,Western blot检测A549细胞中活化型Caspase-3(Cleaved Caspase-3)蛋白表达的变化,同时检测胞浆和线粒体中细胞色素C(Cytochrome C)蛋白表达的变化。以仅用肺炎链球菌诱导的A549细胞为对照。结果:TLR4 siRNA慢病毒可以下调肺炎链球菌诱导的A549细胞中TLR4 mRNA和蛋白的表达水平。肺炎链球菌诱导处理后的A549细胞凋亡率升高,细胞中Cleaved Caspase-3蛋白水平升高,胞浆中Cytochrome C蛋白水平升高,线粒体中Cytochrome C蛋白水平降低。与单纯用肺炎链球菌诱导的A549细胞比较,沉默TLR4后的A549细胞凋亡率降低,细胞中Cleaved Caspase-3蛋白水平降低,胞浆中Cytochrome C蛋白水平降低,线粒体中Cytochrome C蛋白水平升高,差异有统计学意义(P<0.05)。结论:降低TLR4的表达可以抑制肺炎链球菌诱导的A549细胞凋亡,作用机制可能与线粒体凋亡途径有关。
Abstract:
Aim:To study the effects of TLR4 on apoptosis of A549 cells treated by Streptococcus pneumoniae.Methods:A549 cells were infected with TLR4 siRNA lentivirus and treated with Streptococcus pneumoniae, and the silencing effect of TLR4 siRNA on TLR4 expression was detected. Flow cytometry was used to detect cell apoptosis.Western blot was used to detect the expression of Cleaved Caspase-3 protein in cells.At the same time, the expression of Cytochrome C protein in cytoplasm and mitochondria was detected.Results:TLR4 siRNA lentivirus could downregulate the expression level of TLR4 mRNA and protein in A549 cells under pneumococcal conditions. The apoptosis rate of A549 cells treated with Streptococcus pneumoniae increased, the level of Cleaved Caspase-3 protein in the cells increased, and the level of Cytochrome C protein in the cytoplasm increased, and the level of Cytochrome C protein in mitochondria decreased. After silencing TLR4, A549 cells were treated by Streptococcus pneumoniae, the rate of apoptosis reduced, the level of Cleaved Caspase-3 protein in the cells decreased, the level of Cytochrome C protein in the cytoplasm reduced, the level of Cytochrome C protein in mitochondria elevated, compared with A549 cells treated only with Streptococcus pneumoniae, the difference were statistically significant(P<0.05).Conclusion:Streptococcus pneumoniae induces the expression of TLR4 in A549 of alveolar epithelial cells, knocking down its expression can inhibit the apoptosis of A549 cells induced by Streptococcus pneumoniae,the mechanism of action is related to the pathway of mitochondrial apoptosis.
