摘要:
目的:研究阿帕替尼对肺腺癌H1650细胞增殖、活性氧(ROS)水平和凋亡的影响。方法:采用不同浓度(0.000、0.125、0.250、0.500、1.000、2.000、4.000、8.000、16.000、32.000、64.000 μmol/L)的阿帕替尼作用于肺腺癌H1650细胞72 h后,CCK-8法检测细胞增殖抑制率; 平板克隆实验观察0(对照)、2、4、8 μmol/L阿帕替尼作用9 d后H1650细胞的克隆形成率; 0(对照)、8、16、32 μmol/L阿帕替尼处理后,在流式细胞仪上检测H1650细胞内的ROS水平和凋亡情况,采用Western blot方法检测H1650细胞Cleaved Caspase-3蛋白表达情况。结果:阿帕替尼作用后H1650细胞的增殖抑制率升高,组间两两比较,差异均有统计学意义(P<0.05)。与对照组相比,阿帕替尼组H1650细胞克隆形成率降低,凋亡率、细胞内ROS水平、Cleaved Caspase-3蛋白表达升高(P<0.05)。结论:阿帕替尼可抑制H1650细胞增殖,增加细胞内ROS水平,促进凋亡。
Abstract:
Aim:To study the effects of apatinib on proliferation, reactive oxygen species(ROS)level and apoptosis of lung adenocarcinoma H1650 cells.Methods:Cell proliferation inhibition rate was detected by CCK-8 method after H1650 cells were treated by different concentrations(0.000,0.125,0.250,0.500,1.000,2.000,4.000,8.000,16.000,32.000,64.000 μmol/L)of apatinib for 72 hours.Clone experiment was performed to observe the effect of 0,2,4,8 μmol/L apatinib on H1650 cell colony formation.The level of ROS and apoptosis of H1650 cells treated with 0,8,16,32 μmol/L apatinib were detected by flow cytometry. The expression of Cleaved Caspase-3 protein in H1650 cells treated with 0,8,16,32 μmol/L apatinib was detected by Western blot.Results:Apatinib had a concentration-dependent effect on the proliferation of H1650 cells, and could significantly inhibit the colony formation of H1650 cells(P<0.05). Apatinib induced apoptosis of H1650 cells and promoted the level of ROS in H1650 cells(P<0.05). The expression of Cleaved Caspase-3 protein in the apatinib group was higher than that in the control group(P<0.05).Conclusion:Apatinib inhibits the proliferation of H1650 cells, increases the ROS level and induces apoptosis.
