摘要:
目的:研究小干扰RNA(siRNA)降低骨肉瘤MG63细胞中组织蛋白酶B(CB)的表达对细胞增殖、迁移及侵袭的影响。方法:将MG63细胞分成空白对照组、siRNA-2组(转染CB-siRNA-2)和NC组(转染NC-siRNA)3组,分别给予相应的处理后,通过Western blot检测3组细胞中CB蛋白表达的变化,CCK-8、平板单克隆、细胞划痕及Transwell实验检测3组细胞增殖、迁移及侵袭能力的改变。结果:siRNA-2组CB相对表达量低于空白对照组和NC组(P<0.001),细胞克隆形成数、迁移能力和侵袭能力亦明显低于空白对照组和NC组( P<0.001); 与空白对照组相比,siRNA-2组细胞增殖率在培养24、48、72 h后明显下降(P<0.001)。结论:CB-siRNA可有效沉默骨肉瘤MG63细胞中CB的表达,降低MG63细胞的增殖、迁移及侵袭能力。
Abstract:
Aim:To study the changes of proliferation, migration and invasion in osteosarcoma MG63 cells after down-regulating the expression of cathepsin B(CB)in cells.Methods:MG63 cells were divided into blank control group, siRNA-2 group(transfected with CB-siRNA-2)and NC group(transfected with NC-siRNA). After treatment, the expression of CB in cells were detected by Western blot.The proliferation, migration and invasion in 3 groups were detected by CCK-8 and plate monoclonal assay, cell scratches and Transwell assays.Results:Compared with the blank control group and the NC group,the expression of CB in the siRNA-2 group was lower, the number of cell clone formation,invasion ability and migration ability were decreased(P<0.001).Compared with the blank control group,the proliferation rate of the siRNA-2 group was significantly lower at 24, 48, and 72 hour(P<0.001).Conclusion:CB-siRNA can effectively silence the expression of CB and reduce the proliferation, migration and invasion of MG63 cell line.
