摘要:
目的:研究miRNA-210-3p对缺氧诱导海马神经细胞凋亡的影响及机制。方法:将海马神经元HT22细胞分成4组,分别为对照组、缺氧组、阴性对照+缺氧组、miRNA-210-3p+缺氧组,采用qRT-PCR方法检测HT22细胞中miRNA-210-3p的表达,Annexin V-FITC/PI双染法检测细胞凋亡情况,Western blot检测细胞中活化型Caspase-3和Caspase-9蛋白表达水平,用硫代巴比妥酸法检测MDA含量,用二硫代二硝基苯甲酸法检测GSH-Px活性,用水溶性四唑盐法检测SOD活性。结果:转染miRNA-210-3p模拟物的HT22细胞经缺氧处理后,细胞中miRNA-210-3p表达水平升高(P<0.05)。缺氧处理后的HT22细胞凋亡率升高,细胞中活化型Caspase-3和Caspase-9蛋白表达水平升高,GSH-Px、SOD活性降低,MDA含量升高(P<0.05); 上调miRNA-210-3p表达可拮抗缺氧诱导的HT22细胞凋亡和活化型Caspase-3、Caspase-9蛋白表达,提高细胞中GSH-Px、SOD活性,降低细胞中MDA含量(P<0.05)。结论:上调miRNA-210-3p能够抑制缺氧诱导的海马神经细胞凋亡,其作用机制可能与降低氧化应激有关。
Abstract:
Aim:To study the effects and mechanism of miRNA-210-3p on hypoxia induced apoptosis in hippocampal neurons.Methods:Hippocampal neuron HT22 cells were divided into 4 groups:control group,hypoxia group,negative trol+hypoxia group,and miRNA-210-3p+hypoxia group(miRNA-210-3p mimics was transfected into HT22 cells after hypoxia).qRT-PCR was used to detect the expression of miRNA-210-3p, Annexin V-FITC/PI double staining was used to detect apoptosis, Western blot was used to detect the expression level of activated Caspase-3 and Caspase-9 proteins in cells, the content of MDA was detected by thiobarbituric acid, the activity of GSH-Px was detected by dithiodibenzoic acid method, and SOD activity was detected by water-soluble tetrazolium salt method.Results:After being treated with hypoxia,the expression level of miRNA-210-3p in cells transfected by miRNA-210-3p mimics increased.After being treated with hypoxia, the apoptosis rate of HT22 cells increased, the levels of activated Caspase-3 and activated Caspase-9 protein increased, the activity of GSH-Px and SOD decreased, and the content of MDA increased(P<0.05).Up-regulation of the expression of miRNA-210-3p could significantly antagonize the apoptosis of HT22 cells induced by hypoxia and the expressions of activated Caspase-3 and activated Caspase-9 proteins, increase the activity of GSH-Px and SOD in cells, and reduce MDA content in cells(P<0.05).Conclusion:Up-regulation of miRNA-210-3p could inhibit hypoxia-induced apoptosis of hippocampal neurons, and the mechanism is related to the reduction of oxidative stress.
