摘要:
目的:探讨上调miRNA-27a-3p(miR-27a-3p)对人乳腺癌MCF-7细胞增殖、侵袭和迁移能力的影响。方法:将miR-27a-3p模拟物转染至MCF-7细胞中,以转染模拟物对照的细胞为阴性对照,以不转染的细胞为空白对照。通过qRT-PCR检测miR-27a-3p的表达,CCK-8实验检测细胞增殖情况,Transwell实验检测细胞侵袭和迁移能力,Western blot检测细胞中PCNA、MMP-2、MMP-9蛋白的表达。结果:与空白对照组和阴性对照组相比,模拟物组MCF-7细胞中miR-27a-3p表达水平,细胞增殖、侵袭和迁移能力,PCNA、MMP-2和MMP-9蛋白表达水平均升高(P<0.05)。结论:上调miR-27a-3p能够提高MCF-7细胞的增殖、侵袭和迁移能力,其机制可能与促进PCNA、MMP-2和MMP-9蛋白表达有关。
Abstract:
Aim:To investigate the effects of up-regulation of miRNA-27a-3p(miR-27a-3p)on proliferation, invasion and migration of human breast cancer MCF-7 cells.Methods:MCF-7 cells were cultured in vitro, and miR-27a-3p mimics were transfected into MCF-7 cells. The cells transfected mimics control were used as negative control, and the non-transfected cells were used as blank control. qRT-PCR was used to detect transfection efficiency. CCK-8 assay was used to detect the proliferation of MCF-7 cells. Transwell assay was used to detect the invasion and migration of MCF-7 cells. Western blot was used to detect the expressions of PCNA, MMP-2, and MMP-9 protein in MCF-7 cells.Results:Compared with the negative control group and the blank control group, miR-27a-3p expression level was significantly increased in miR-27a-3p mimics group, cell proliferation, invasion and migration were significantly prompted, and PCNA, MMP-2 and MMP-9 protein levels were significantly increased(P<0.05).Conclusion:Up-regulation of miR-27a-3p could promote the proliferation, invasion and migration of MCF-7 cells, which may be related with its promoting the expressions of PCNA, MMP-2 and MMP-9 proteins.
