摘要:
目的:探讨上调血小板凝血酶蛋白1(THBS1)的表达对肺腺癌H1975细胞体外迁移能力的影响。方法:H1975细胞分为3组,空白对照组不处理,空载体转染组及pcDNA-THBS1转染组分别用脂质体转染空载体pcDNA3.1和pcDNA3.1-THBS1真核表达载体,采用划痕实验检测细胞迁移能力,用qRT-PCR、Western blot检测细胞中基质金属蛋白酶(MMP)-2和MMP-9 mRNA、蛋白的表达情况。结果:与空白对照组和空载体转染组相比,pcDNA-THBS1转染组H1975细胞迁移能力增强,细胞中MMP-2和MMP-9 mRNA和蛋白的表达升高(P<0.05)。结论:上调THBS1蛋白可以增强肺腺癌H1975细胞体外迁移能力。
Abstract:
Aim:To study the effects of thrombospondin 1(THBS1)up-regulation on the migration ability of lung adenocarcinoma cells in vitro.Methods:H1975 cells were allocated into 3 groups,blank control group was not treated,and empty vector transfection group and pcDNA-THBS1 transfection group were transfected with empty vector and pcDNA3.1-THBS1 eukaryotic expression vector by liposome. The migration ability of cells was detected by cell scratch test. The mRNA and protein expressions of matrix metalloproteinase-2(MMP-2)and matrix metalloproteinase-9(MMP-9)were detected by qRT-PCR and Western blot.Results:Compared with blank control group and empty vector transfection group, the migration ability of cells in pcDNA-THBS1 transfection group was significantly enhanced, and the expressions of MMP-2 and MMP-9 mRNA and protein in the cells were significantly increased(P<0.05).Conclusion:Increasing the expression of THBS1 protein enhances the migration ability of H1975 cells in vitro.
