摘要:
目的:研究夏天无总碱对血管性痴呆大鼠海马细胞凋亡及认知功能的影响。方法:50只大鼠分为对照组,模型组和0.3、0.6和1.2 mg/kg夏天无总碱组,每组10只。模型组和夏天无总碱组构建血管性痴呆大鼠模型,0.3、0.6和1.2夏天无总碱组分别灌服0.3、0.6和1.2 mg/kg的夏天无总碱30 d。Morris水迷宫实验检测大鼠认知能力,用相应试剂盒分别检测大鼠海马区去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)、乙酰胆碱酯酶(AchE)、丙二醛(MDA)含量和超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,TUNEL法检测海马神经元凋亡情况,Western blot检测活化型Caspase-3(c-Caspase-3)蛋白表达水平。结果:模型组大鼠认知能力降低,NE、DA、5-HT、AchE含量下降,神经元凋亡率升高,c-Caspase-3蛋白水平升高,SOD、GSH-Px活性降低,MDA含量升高(P均<0.05)。夏天无总碱组大鼠认知能力改善,NE、DA、5-HT、AchE含量升高,神经元细胞凋亡率降低,c-Caspase-3蛋白水平降低,SOD、GSH-Px活性升高,MDA含量下降(P均<0.05)。结论:夏天无总碱能够改善血管性痴呆大鼠认知能力,其作用机制可能与抑制氧化应激、减少海马神经元细胞凋亡、调节神经递质释放有关。
Abstract:
Aim:To study the effects of total alkaloids of Rhizoma Corydalis on the apoptosis of hippocampal cells and cognitive function in vascular dementia rats.Methods:A total of 50 rats were allocated into control group, model group and 0.3, 0.6 and 1.2 mg/kg total alkaloids of Rhizoma Corydalis groups, 10 rats in each group. Vascular dementia rat models were established in model group and total alkaloids of Rhizoma Corydalis groups. The 0.3, 0.6 and 1.2 mg/kg total alkaloids of Rhizoma Corydalis groups were administered with corresponding concentration of total alkaloids of Rhizoma Corydalis for 30 days, respectively.Morris water maze test was used to detect the cognitive ability of rats.The levels of NE, DA, 5-HT, AchE, SOD, GSH-Px and MDA in hippocampus of rats were detected by kits.TUNEL method was used to detect apoptosis of hippocampal neurons.The expression level of c-Caspase-3 protein was detected by Western blot.Results:The cognitive ability of vascular dementia rats was significantly reduced, the levels of NE, DA, 5-HT and AchE decreased, the apoptosis rate of neurons increased, the level of c-Caspase-3 protein increased, the activity of SOD and GSH-Px decreased, and the content of MDA increased(P<0.05).After the use of total alkaloids of Rhizoma Corydalis,the cognitive ability of vascular dementia rats was obviously improved, the contents of NE, DA, 5-HT and AchE increased, the apoptosis rate of neurons decreased, the level of c-Caspase-3 protein decreased, the activity of SOD and GSH-Px increased,and the content of MDA decreased(P<0.05).Conclusion:Total alkaloids of Rhizoma Corydalis can improve the cognitive function of vascular dementia rats,and its mechanism may be related with inhibiting oxidative stress, reducing apoptosis of hippocampal neurons and regulating neurotransmitter release.
